摘  要:目的探索miR-671在胃癌组织和细胞中的表达,并研究其对胃癌细胞增殖和迁移的影响。方法通过qRT-PCR实验检测miR-671在胃癌组织和细胞中的表达,通过CCK-8实验检测miR-671表达上调对MKN45胃癌细胞增殖,通过划痕和Transwell小室实验检测miR-671表达上调对MKN45胃癌细胞侵袭和迁移的影响。结果与癌旁正常组织相比,胃癌组织中的miR-671表达明显下调(P<0.01);与正常人胃黏膜上皮细胞相比,胃癌细胞中的miR-671表达也明显下调(P<0.01);miR-671表达上调明显抑制MKN45胃癌细胞增殖、迁移和侵袭(P<0.01)。结论 miR-671可以抑制胃癌细胞的增殖、迁移和侵袭,在胃癌中发挥抑癌基因功能。Objective To investigate the expression of miR-671 in gastric cancer tissues and cell lines, and determine its effects on the proliferation and invasion of gastric cancer cells. Methods The expression of miR-671 was detected in gastric cancer tissues and para-carcerous tissues from 8 patients and cell lines SGC7901, MKN28 and MKN45, and human gastric mucosa] epithelial cell line GES-1 by quantitative real-time PCR (qRT-PCR). CCK-8 assay was used to measure the cell proliferation after miR-671 up- regulation. The effect of miR-671 on cell migration and invasion was detected by wound healing assay and Transwell chamber assay. Results The expression level of miR-671 was significantly decreased in gastric cancer tissues than the adjacent normal tissues (P 〈 0. 01 ). The expression levels of miR-671 were significantly decreased in the gastric cancer cell lines compared with GES-1 cells ( P 〈 0. 01 ). Up-regulation of miR-671 significantly inhibited the proliferation, migration and invasion in gastric cancer cells (P 〈 0. 01 ). Conclusion miR-671 inhibits proliferation, migration and invasion of gastric cancer cells, and may serve as a tumor suppressor.
