补肾活血固齿方对MC3T3-E1细胞矿化结节形成影响的研究

摘　　要：目的观察补肾活血固齿方对小鼠颅顶前成骨细胞(MC3T3-E1细胞)矿化结节形成的影响,探讨该方剂促进成骨的作用机制。方法依照体表面积折算成等效剂量的补肾活血固齿方灌胃大鼠,制备含药血清(含药血清组);同等剂量的蒸馏水灌胃大鼠,制备无药血清(无药血清组);胎牛血清作为空白对照(胎牛血清组)。MC3T3-E1细胞分别于补肾活血固齿方含药血清、无药血清、胎牛血清中培养14 d,利用茜素红染色法观察各组矿化结节的形成情况并计数。结果 3组细胞在14 d时均形成矿化结节,含药血清组矿化结节形成数量明显多于无药血清组及胎牛血清组(P均<0.05),无药血清与胎牛血清组矿化结节形成数量比较差异无统计学意义(P>0.05)。结论补肾活血固齿方可促进MC3T3-E1细胞向成骨细胞分化成熟,通过促进牙周骨组织的再生而发挥其药理作用。Objective It is to observe the effect of Bushen Huoxue Guchi decoction on mineralized nodule formed in MC3T3-E1 cells, so that to explore the mechanism of this decoction on osteogenesis. Methods Contained-herb serum as contained serum group was prepared from rats fed with Bushen Huoxue Guchi decoction according with body surface area converted equivalent dose by lavage; blank-contained serum as blank-contained serum group was prepared with normal saline at the same time, and fetal bovine serum （FBS） as blank control group. MC3T3-Elcells were cultured in contained-herb serum,no- contained serum, and FBS respectively for 14 days. The mineralized nodules formation in every group were evaluated by alizarin red staining. Results Mineralized nodules were formed in all groups after cultivated for 14 days. The average volume of miner-alized nodules were larger in contained-herb serum than that in blank-contained serum and blank controlled groups （P 〈0. 05）, there was no difference for statistical significance between blank-contained serum and blank control groups（F 〉0. 05）. Conclusion Bushen Huoxue Guchi can accelerate the differentiation and maturation of MC3T3 - El cells, and promote the regeneration of alveolar bone to play its pharmacological action.Key words
