摘 要:目的:研究hsa-mi R-127-3p(mi R-127)在宫颈癌组织中的表达水平及其与临床病理特征之间的相关性,探讨mi R-127对宫颈癌细胞株Hela及Siha迁移和侵袭的影响。方法:Real-time PCR法检测50对宫颈癌及癌旁组织中mi R-127表达水平,分析mi R-127表达与临床病理特征的相关性。细胞转染mi R-127 mimics及mimics negative control,采用划痕愈合实验检测细胞迁移能力,Transwell法检测细胞侵袭能力。结果:mi R-127在宫颈癌组织中的表达明显低于癌旁组织(P<0.05)。mi R-127表达与淋巴脉管间隙浸润相关(P=0.048),而与年龄、FIGO分期、病理类型、分化程度、病灶大小、浸润深度、有无淋巴结转移无关(P>0.05)。过表达mi R-127可抑制Hela及Siha细胞的迁移和侵袭能力。结论:mi R127在宫颈癌组织中低表达,并且抑制宫颈癌细胞株Hela及Siha的迁移和侵袭,可能与宫颈癌的发生发展密切相关。Objective:To investigate the expression level of hsa-miR-127-3p(miR-127)in cervical cancer and it''s relevance to the clinical pathology features of cervical cancer,besides,to detect the effect of miR-127 on the migration and invasion of Hela and Siha cells.Methods:The miR-127 expression was detected by real-time PCR in 50 cases of cervical cancer tissues and paired normal tissues.After the cell 1ines were transfected with miR-127 mimics and miR-127 mimics negative control.The Wound-healing assay was used to observe the cell migration,the transwell assay was used to determine cell invasion.Results:The expression of miR-127 in cervical cancer tissues was lower than that in the paired normal tissues (P〈0.05).The expression of miR-127 in patients with lymph-vascular space invasion was lower than that with no lymph-vascular space invasion (P=0.048),but there was no correlation with age,FIGO stage,pathology type,degree of differentiation,invasion depth and lymph node metastasis.Overexpression of miR-127 inhibits migration and invasion in Hela and Siha cells.Conclusion:miR-127 is low-expressed in cervical cancer and inhibits the migration and invasion in cervical cancer cell lines.miR-127 might be associated with the genesis and development of cervical cancer.
