摘  要:制备了微米级甲基丙烯酸β-羟乙酯和N-乙烯基吡咯烷酮聚合物微球HEMA/NVP,再利用微球羟基与环氧氯丙烷发生亲核取代反应将环氧基引入微球表面,制得了表面含有环氧基的固定化酶载体EH-HEMA/NVP.利用红外光谱和化学分析法对载体进行表征,并考察载体对青霉素酰化酶的固定化效果.研究结果表明:载体EH-HEMA/NVP的溶胀性能优越,能较好的保持酶的活性;亲核取代反应时间会影响载体表面环氧基的含量;该载体对酶的固定较为牢固,重复使用14次以上仍然保持较高的相对活性.The micron--sized copolymer microspheres were prepared by hydroxyethyl methacrylate and N --vinylpyrrolidone. Oxirane was then brought into the surface of microspheres via the nucleophile substitution reaction between the hydroxyl groups and epichlorohydrin, to form carrier EH--HEMA/NVP. The carrier was characterized by FTIR and chemical analysis methods; the immobilized effect on penicillin was examined. The results indicate that the carrier displays better swelling capacity and constellation and activity of enzyme was excellent maintained; the reaction time has great effect on the content of oxirane; the enzyme can stably combined with the carrier, and its relative activity can keep a high level after repeatedly operation for 14 times.
