摘  要:目的探讨青蒿琥酯(ART)对结肠癌细胞增殖作用及其可能机制。方法人结肠癌细胞(HCT116)常规培养,分对照组、ART低剂量组(10μg/mL)和ART高剂量组(20μg/mL),绘制生长曲线检测相关蛋白。siPRMT6转染细胞,Real time PCR和Western blot检测ART对结肠癌细胞增殖及相关蛋白表达的影响。结果 HCT116细胞经10和20μg/ml ART处理后72 h,细胞增殖显著降低,p27和Cyclin D1表达水平显著降低,p21蛋白显著升高。PRMT6基因沉默后,ART不能发挥作用,逆转p27、Cyclin D1和p21 mRNA和蛋白表达变化。结论 ART能显著抑制结肠癌细胞增殖,可能与p27、Cyclin D1和PRMT6蛋白有关。Objective To investigate the effect of artesunate on proliferation of colon cancer cells and its possible mechanism. Methods Human colon cancer cells (HCT116) were cultured and divided into control group, ART low dose group(10g/mL) and ART high dose group(20g/mL), the growth curve was drawn to detect related proteins. The siPRMT6 was transfected into HCT116 ceils, Real time PCR and Western blot were used to detect the effect of ART on the proliferation of colon cancer and the expressions of related proteins. Results HCT116 cells were treated with 10 and 20~lg/ml ART g/ml for 72 h, the cell proliferation and p27 and Cyclin D1 expression level was significantly reduced, but p21 protein was significantly increased. PRMT6 gene silencing, reversed the expressions of p27, Cyclin and p21 mRNA and protein. Conclusion The inhibition of ART the proliferation of colon cancer cells might be related to the expression of p27, Cyclin D1 and PRMT6 proteins.
