摘 要:目的探讨微小RNA-191(miR191)调控SH3结构域结合蛋白5(SH3BP5)基因对三阴性乳腺癌患者预后的影响。方法收集新疆医科大学附属肿瘤医院2010年1月至2015年1月收治的216例手术切除并经病理学诊断的三阴性乳腺癌患者乳腺组织废弃标本和完整临床资料,选取健康女性(乳腺癌疑似患者活检后正常)正常乳腺组织样本5例作为对照。采用实时定量聚合酶链反应检测miR191、SH3BP5 mRNA表达,采用蛋白质印迹法检测其蛋白表达,分析miR191、SH3BP5与三阴性乳腺癌患者预后的关系。采用蛋白质印迹法检测三阴性乳腺癌组织凋亡蛋白——PARP剪切体表达情况。结果三阴性乳腺癌组织miR191 mRNA表达(3.28±1.75)明显高于正常乳腺组织(0.73±0.32),SH3BP5 m RNA表达(0.79±0.24)明显低于正常乳腺组织(3.67±0.31),差异均有统计学意义(P<0.05);三阴性乳腺组织SH3BP5蛋白表达水平也明显低于正常乳腺组织。SH3BP5表达水平高的患者5年生存率[88.43%(191/216)]高于SH3BP5表达水平低者[50.46%(109/216)]。SH3BP5表达水平低的患者PARP剪切体表达升高。结论 miR191抑制SH3BP5表达是三阴性乳腺癌患者预后不良的标志。Objective To explore the prognostic effect of miR191-SH3BP5 in triple negative breast cancer patients.Methods The breast tissue discarded specimens and complete clinical data of 216 patients with triple negative breast cancer who performed surgery resection and the pathological diagnosis in the Afflicated Hospital of Xinjiang Medical University from January 2010 to January 2015 were collected. Five specimens from healthy women who were diagnosed with suspected breast cancer but normal tissue biopsies were taken as the control. qRT-PCR was used to detected the miR191 and m RNA expression of SH3BP5 while Western blotting was used to evaluated the protein expression level. The correlation between miR191,SH3BP5 and the prognosis of triple negative breast cancer was analysed. Western blotting was applied to detected the expression level of apoptosis protein PARP spliced variants of triple negative breast cancer. Results Compared with normal female breast tissue(0.73±0.32),the miR191 expression level of triple negative breast cancer patients(3.28±1.75) was higher,the SH3BP5 m RNA and protein expression level of normal female breast tissue(0.79 ±0.24) was lower than that of triple negative breast cancer( 3. 67 ± 0. 31), the difference was statistically significant( P 0. 05). The 5-year survival rate [88.43%(191/216)] of SH3BP5 high expression patients was higher than that of low SH3BP5 expression patients [50.46%(109/216)]. Low SH3BP5 expression patients had an elevated expression level of PARP spliced variants. Conclusion Mi R191 inhibits SH3BP5 expression as a sign of poor prognosis in patients with triple negative breast cancer.
