摘 要:目的研究赖氨酰氧化酶(LOX)基因G473A多态性与结直肠癌易感性的关系,为结直肠癌肿瘤的风险评估提供依据。方法随机选择2010年5月~2014年8月我院收治的200例结直肠癌患者作为研究对象,并随机选择同期200例体检健康者作为对照组,分别抽取所有研究对象的肘静脉血5 mL和抽取基因组后对LOX基因G473A位点测序,检测各个体LOX基因G473A的基因型。然后采用Logistic回归分析不同LOX基因G473A基因型与结直肠癌患病风险之间的相关性。结果 (1)结肠癌组GG、GA、AA基因型患者分别占47.37%(45/95)、41.05%(39/95)、11.58%(11/95)。携带GG基因型个体患结肠癌的风险明显低于GA和AA基因突变型个体(OR=1.71,95%CI=1.01~2.65,P=0.03;OR=3.94,95%CI=1.43~8.96,P=0.00)。(2)直肠癌组GG、GA、AA基因型患者分别占51.43%(54/105)、37.4%(39/105)、11.43%(12/105)。携带GG基因型个体患直肠癌的风险明显低于AA基因突变型个体(OR=3.45,95%CI=1.61~8.03,P=0.00)。(3)结直肠癌组GG、GA、AA基因型患者分别占49.50%(99/200)、39.00%(78/200)、11.50%(23/200)。携带GG基因型个体患结直肠癌的风险明显低于GA和AA基因突变型个体(OR=1.67,95%CI=1.01~2.18,P=0.03;OR=3.68,95%CI=1.81~7.65,P=0.00)。结论 LOX基因G473A多态性与结直肠癌的发病风险有一定关系,行LOX基因G473A多态性检测在结直肠癌风险评估中具有一定的应用价值。Objective To study the relationship between the G473A polymorphism of lysyl oxidase(LOX) gene and the susceptibility of colorectal cancer, and to provide the basis for the risk assessment of colorectal cancer. Methods A to- tal of 200 patients with colorectal cancer were enrolled in our hospital from May 2010 to August 2014.A total of 200 cases of normal physical examination were recruited as control group. All the subjects were given 5 mL of elbow vein blood. Genomic DNA was extracted from peripheral blood and the G473A locus of LOX gene was sequenced and the genotype was detected. Logistic regression was used to analyze the correlation between the G473A genotype of LOX gene and the risk of colorectal cancer. Results (1)The proportion of GG, GA and AA genotype in eolorectal cancer group were 47.37% (45/95), 41.05% (39/95)and 11.58% (11/95) respectively. The risk of colorectal cancer with GG genotype was significantly lower than that of GA and AA mutant individuals(OR=1.71, 95%CI=1.01~2.65, P=0.03; OR=3.94, 95%CI=1.43-8.96, P=0.00).(2)Tbe genotype of GG, GA and AA in rectal cancer group was 51.43%(54/105), 37.4%(39/105) and 11.43%(12/105) respectively. The risk of rectal cancer with GG genotype was significantly lower than in AA mutant individuals(OR=3.45, 95%CI=1.61-8.03, P=0.00).(3)The proportion of GG, CA and AA genotype in colorectal cancer group was 49.50%(99/200), 39.00%(78/200) and 11.50%(23/200) respectively. The risk of col- orectal cancer with GG genotype was significantly lower than that of GA and AA mutant individuals(OR=1.67, 95%CI= 1.01-2.18, P=0.03; OR=3.68, 95%CI=1.81~7.65, P=0.00). Conclusion The G473A polymorphism of LOX gene is associated with the risk of colorectal cancer. The detection of LOX gene G473A polymorphism has a certain value in the risk assessment of colorectal cancer.
