结直肠癌组织乳脂肪球表皮生长因子8蛋白表达与临床病理特征和患者生存率的关系

摘　　要：目的:探讨乳脂肪球表皮生长因子8(MFG-E8)蛋白在结直肠癌(CRC)组织中的表达及其与临床病理特征和患者生存率的关系。方法:留取手术切除CRC癌组织(CRC组)及相应癌旁组织(Control组)各88例,应用免疫组织化学法(SP法)检测癌组织及癌旁组织MFG-E8蛋白表达,统计分析各组MFG-E8蛋白高表达率的差异,以及CRC组中MFG-E8低表达组、阳性表达组和强阳性表达组患者临床病理因素和总生存率的差异。结果:CRC组癌组织MFG-E8蛋白高阳性表达率为73.86%(65/88),显著高于Control组28.41%(25/88)(P<0.01)。MFG-E8蛋白表达强度仅与淋巴结转移有关(P<0.05),而与患者年龄、临床T和M分类、临床分期及肿瘤分型无显著相关(P>0.05);MFG-E8阴性表达组及强阳性表达组患者总生存率均显著低于阳性表达组(P<0.01)。结论:MFG-E8在CRC患者癌组织中阴性表达及强阳性表达均可促进CRC的淋巴结转移,并提示预后较差。Objective：To explore the expression of milk fat globule EGF factor Ⅷ（MFG-E8）in colorectal cancer（CRC）tissues and the correlation with clinicopathological features and survival in CRC patients.Method：88CRC-related cases with follow-up information were made into tissue microarrays according to the paired principle of cancer tissues and the adjacent tissues.The expression of MFG-E8 was detected with immunohistochemical analysis and SPSS software was finally utilized to analyze the relationships between experimental data and clinical indicatives.Results：MFG-E8 expression increased significantly in colorectal cancer compared with normal mucosa tissues（control group）（P〈0.01）.In addition,both strong positive and negative expression of MFG-E8 were correlated with lymph node metastasis（P〈0.05）.Furthermore,patients with strong positive or negative expression of MFG-E8 showed shortened overall survivals（P〈0.01）,compared with patients with weakly positive and positive expression of MFG-E8.Conclusion：Both strong positive expression and negative expression of MFG-E8 might be involved in the progression of CRC and they were associated with patients＇ poor overall survivals.
