摘 要:目的建立了一种血液中氰化物的电化学检测器/离子色谱检测方法。方法使用甲醇沉淀血液中的血红蛋白,经高速离心后,通过活化Dionex On GuardⅡRP过滤柱,采用美国戴安ICS-5000型离子色谱仪配以AS19高容量色谱柱,20 mmol/L KOH全自动淋洗液发生器,Ag/Ag Cl三电极体系电解池,ED电化学检测器,对氰离子进行检测,建立了一种快速、准确的测定方法。结果实验表明氰离子在0.005~5μg/m L浓度范围内有良好的线性关系(r^2=0.99968),依据S/N=3计算方法的检出限为1×10^(-4)μg/m L,依据S/N=10方法的定量下限为4×10^(-4)μg/m L,CN-的空白加标回收率为84%~120%,相对标准偏差(RSD)为3.18%。结论实验表明本方法前处理过程简单,方法检出限低、灵敏度高、操作简单,适用于血液中氰离子含量的测定。Objective A method was established to determine cyanide ion in human blood based on ion chromatography.Method 3m L of methanol was added to 1m L of heart blood to precipitate the protein in blood. The sample was centrifuged for 5min at the speed of 10,000 rpm. The supernatant was filtered through activated Dionex On Guard Ⅱ RP column and separated on an Ion Pac AS19 column with the 20mmol/L KOH solution. The eluent was detected by an ED electro chemical detector with an Ag/Ag Cl three electrode system for cell. Results The limit of detection(LOD) of CN-is 1×10^-4μg/m L. The limit of quantification(LOQ) was 4×10^-4μg/m L. In the range of 0.005~5μg/m L, it has a good linear relationship(r^2=0.99968).The recovery of spiked samples was 84%-120%, with a relative standard deviation(RSD) of 3.18%. Conclusion The sample preparation is simple. The established method is sensitive and can be used to determine cyanide ion in human blood for related poisoning cases.
