摘 要:目的:探讨沉香醇提物对四氯化碳(CCl4)致小鼠急性肝损伤的保护作用。方法:采用CCl4腹腔注射制备小鼠急性肝损伤模型,给药处理后计算肝脏指数;检测血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)活性;肝组织匀浆液中髓过氧化物酶(MPO)、一氧化氮(NO)、总抗氧化能力(T-AOC)和超氧化物歧化酶(SOD);肝脏进行病理切片观察;酶联免疫法(ELISA)检测血清中白介素-1β(IL-1β)、白介素-10(IL-10)的表达水平。结果:与模型组相比,通体沉香醇提物能剂量依赖性显著降低血清ALT、AST活性,降低MPO活性、NO含量,提高T-AOC能力及SOD活性;缓解肝脏组织病理损伤;减低IL-1β和升高IL-10的表达水平,且通体沉香醇提物高剂量效果最好,优于等剂量的野生沉香和火烙沉香醇提物。结论:沉香醇提物对肝损伤有保护作用,其机制可能与抗氧化应激,抑制脂质过氧化及抗炎作用有关。Objective:This study is aimed to investigate the hepatoprotective effect of alcohol extract of agarwood on acute liver injury induced by carbon tetrachloride (CCl4) in mice.Methods:Mice were injected with CCl4 to establish acute liver injury model.Alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were measured of serum.The myeloperoxidase (MPO),nitric oxide (NO),total peroxide (T-AOC) and superoxide dismutase (SOD) of the liver tissue homogenate were measured,and observed degree of liver injury by pathological sections.The interleukin-1β (IL-1β) and Interleukin-10 (IL-10) expression levels were measured by enzyme-linked immunosorbent assay (ELISA).Results:Compared with the model group,the alcohol extract of Agar-Wit agarwood groups significantly decreased the activities of serum ALT,AST,MPO and NO contents as well as the expression level of IL-1β,enhanced the capacity of T-AOC,the activity of SOD and IL-10 level,and improved pathological injury of liver tissue,especially the alcohol extract of Agar-Wit agarwood 2.84 g/kg group.Conclusion:Alcohol extracts of agarwood show a significantly protective effect on acute liver injury in mice,and its mechanism may be related to anti-oxidative stress,inhibition of lipid peroxidation and anti-inflammatory effect.
