摘 要:细胞培养技术是生物技术中最核心、最基础的技术,目前常用的二维细胞培养方法,例如,多孔板、烧瓶和培养皿等存在诸多不足和限制,三维细胞培养能够改善细胞局部微环境、促进细胞与细胞、细胞与基质之间的交流,模拟体内生理环境,从而增强细胞功能,在新药研发、肿瘤研究等领域具有广泛应用前景。支架是实现三维细胞培养最常用的手段之一,相当于人工细胞外基质,能够模拟体内组织复杂的三维结构及其主要特征。本文从基于支架的培养体系和无支架的培养体系两方面讨论了三维细胞培养方法,尤其详细介绍了各种支架。Cell culture technology is one of the most core and basic technologies in biotechnology. At present,the limitation and shortcomings of two-dimensional cell culture methods such as multi-well plates,flasks and Petri dishes are obvious. Three-dimensional cell culture can improve the local microenvironment,cell-cell and cell-extracellular matrix interactions,mimic the physiological conditions in vivo to enhance the cell function. Three-dimensional cell culture might be widely used in oncology,drug discovery and other fields in the future. Scaffold is one of the most popular methods for three-dimensional cell culture,which is equivalent to artificial extracellular matrix to mimic the complex three-dimensional structure and the main characteristics of living tissues. This review summarizes three-dimensional cell culture from two aspects: scaffold-based and scaffold-free three-dimensional cell culture,especially introduces the scaffolds.
