摘 要:本实验旨在研究红桂木凝集素(Artocarpus lingnanensis lectin,ALL)对人鼻咽鳞癌细胞株CNE1和肝腹水腺癌细胞株SK-Hep-1增殖的影响。利用亲和层析纯化红桂木凝集素;不同浓度ALL处理CNE1和SK-Hep-1细胞48 h后,倒置显微镜观察ALL对CNE1和SK-Hep-1细胞形态的影响,CCK8法检测不同浓度ALL对CNE1和SK-Hep-1增殖的影响。经纯化的红桂木凝集素,凝集红细胞活力为:29~211。倒置显微镜观察发现,随着ALL浓度的增加,细胞形态变小且皱缩,透光性差,出现细胞碎片。CCK8法表明,低剂量ALL对细胞无抑制作用,高剂量ALL明显抑制CNE1和SK-Hep-1细胞的增殖,呈剂量-浓度依赖关系。由此得出高浓度ALL能抑制鼻咽癌细胞株CNE1,肝癌细胞株SK-Hep-1的增殖的结论。This experiment aimed at studying the effect ofA rtocarpus lingnanensis lectin (ALL) on the proliferation of nasopharyngeal squamous carcinomas cells line CNE 1 and liver ascites gland cancer cells line SK-Hep-1. Affinity chromatography was used to purify A rtocarpus lingnanensis lectin from its seeds. CNE1, SK-Hep-1 were incubated with different concentrations of ALL for 48 h. The effect of ALL on cells morphology and cell prolifera- tion of CNE 1 and SK-Hep-1 was observed by inverted microscope and analyzed by CCK8 method, respectively. The agglutinating activity of purified ALL was 29-21l. The observation of inverted microscope showed that with the increase of the concentration of ALL, the morphology of cells was changed, showing cell shrinkage, decreased transparency as well as the appearance of cell debris. CCK8 assay indicated that ALL with low concentration had no inhibitory effects on cell growth, while ALL with high concentration had obvious inhibitory effects on the pro- liferation of CNE1, SK-Hep-1 cells, with a dose-concentration independent manner. We would be able to make a conclusion that high concentration of ALL could inhibit the proliferation of CNE 1 and SK-Hep-1 cell lines.
