摘 要:目的探讨牙移植术后牙周组织损伤修复中人脐带间充质干细胞应用的效果。方法将实验小鼠随机均分为两组,选取细胞量需大于10~6个的样品作为实验组,再选取未经诱导的细胞作为对照组。实验组材料(牙本质复合体)植入于左侧,对照组材料(牙本质复合体)植入于右侧,同样条件饲养小鼠8周后取材。采用矿化液进行成骨诱导,检测细胞的成骨活性;然后将培养7 d以后的复合膜分别移植到牙周组织损伤处,8周以后对组织学染色牙周再生进行检测,观察其生长情况、Masson三色染色的骨样结构以及细胞、生物蛋白胶以及牙本质复合体,同时观察手术4周后血清碱性成纤维细胞因子(b EGF)、转化生长因子-β1(TGF-β1)、骨形态发生蛋白-7(BMP-7)指标水平。结果实验组成骨活性更加明显,且其成骨的分化需要一个连续的过程,相比对照组,实验组更适用于骨缺损的修复。手术后4周,实验组血清b EGF、TGF-β1、BMP-7水平明显高于对照组,差异具有统计学意义(P<0.05)。HE、Masson染色、免疫组化均可见,诱导后的人脐带间充质干细胞牙本质复合体周围损伤组织恢复更迅速,形成大量胶原纤维或类骨样结构更明显。结论经过诱导的人脐带间充质干细胞可以有效改善血清b EGF、TGF-β1、BMP-7水平,更适用于牙周损伤的修复。Objective To investigate the effect of human umbilical cord mesenchymal stem cells applications in the dental periodontal tissue repairment after transplantation. Methods The experimental mice were randomly divided into two groups. A sample with more than 10-6 cells was selected as the experimental group,and the untreated cells were selected as the control group. The experimental group( dentin complex) was implanted on the left side. The control group was implanted on the right side and the mice were given the same condition for 8 weeks. The separation between the in vitro human umbilical cord mesenchymal stem cells. The use of liquid mineralized osteogenic detecting osteogenic activity of the cells. And then cultured for 7 days after composite membranes were transplanted into periodontal tissue lesions after eight weeks of histological staining periodontal regeneration were detected to observe the growth,Masson trichrome bone-like structure and the cells,fibrin glue and dentin.The levels of serum b EGF,TGF-β1 and BMP-7 were observed at 4 weeks after operation. Results Between human umbilical cord induced mesenchymal stem cell osteogenic activity more obvious,and the umbilical cord differentiation of mesenchymal stem cells into bone requires a continuous process,inter-after-induced human umbilical cord mesenchymal stem cells are more suitable than no induced cell bone defect. At 4weeks after operation,the levels of serum b EGF,TGF-β1 and BMP-7 in the observation group were significantly higher than those in the control group. The levels of serum b EGF,TGF-β1 and BMP-7 were statistically significant( P〈0. 05). HE,Masson staining,immunohistochemistry were visible,human umbilical cord mesenchymal stem cells induced by human umbilical cord mesenchymal stem cells were recovered more rapidly,and the formation of a large number of collagen fibers or bone like structure was more obvious.
