干蟾皮提取物对胃癌肝转移裸鼠CXCL12-CXCR4轴的影响

摘　　要：目的:观察干蟾皮提取物(华蟾素注射液)对裸鼠胃癌肝转移及CXCL12-CXCR4生物轴的影响。方法:MTT法测定华蟾素注射液对人胃癌BGC-823细胞体外增殖的抑制作用;经小鼠脾脏下极包膜注入BGC-823细胞建立胃癌肝转移模型;32只小鼠随机分为模型组、华蟾素低剂量组[CINO(低)]、华蟾素中剂量组[CINO(中)]、华蟾素高剂量组[CINO(高)];给予相应药物干预,观察不同浓度的华蟾素注射液对BGC-823细胞的抑制作用及对胃癌肝转移裸鼠生存时间、肿瘤抑制率,肿瘤组织中CXCL12、CXCR4的影响。结果:华蟾素对BGC-823细胞的增殖具有抑制作用;CINO高剂量组较模型组肝转移率明显下降(P<0.05);华蟾素注射液对胃癌肝转移组织中CXCL12和CXCR4蛋白的表达均有一定的抑制作用(P<0.05)。结论:华蟾素注射液可抑制BGC-823细胞的增殖,降低胃癌肝转移率,抑制肿瘤生长;其机理可能与其下调胃癌肝转移灶中CXCL12、CXCR4蛋白表达有关。Objective： To observe the effects of dry toad skin extracts on CXCL12-CXCR4 axis in nude mice with liver metastasis of gastric cancer. Methods： The model of nude mice were established by using BGC-823 cells. And then the 32 model mice were interferenced with model group,Huachansu low dose group [CINO（ low） ],Huachansu middle dose group [CINO（ middle） ],Huachansu high dose group [CINO（ high） ]. After the intervention,observe the influence of them on nude mice model of liver metastasis of gastric cancer,the degree of liver metastases and CXCL12 and CXCR4. Results： Huachansu has certain inhibitory effect on liver metastasis of gastric cancer of nude mice,CXCL12 and CXCR4 gene expressions in the tissue（ P〈0. 05）. Huachansu high dose group can enhance the inhibitory effect of liver metastasis of gastric cancer of nude mice than model group. Huachansu dose groups all had effect on the CXCL12 and CXCR4 gene expressions in the tissue（ P〈0. 05）. Conclusion： Huachansu high dose group can enhance the inhibitory effect of BGC-823 cells,reduce liver metastasis of gastric cancer of nude mice,and finally suppress the tumor growth. The mechanism may be associated with Huachansu enhancing the efficacy of liver metastasis of gastric cancer of CXCL12 and CXCR4 gene expressions.
