摘　　要：目的:研究肿瘤相关基因NGX6在胃癌与大肠癌组织以及其配对癌旁正常组织中的表达,探讨此基因在胃癌、大肠癌发生发展中的作用.方法:采用RT-PCR方法、点杂交、Northern-blot方法分别检测34例胃癌、34例大肠癌患者癌瘤组织、癌旁(手术切缘,距肿瘤5cm以上)正常组织NGX6基因表达.结果:RT-PCR显示:NGX6基因在73.5%的大肠癌组织(25/34)和93.8%的有淋巴结或远处转移的大肠癌组织(15/16)中存在表达减弱或不表达,其表达下调率明显高于相应的癌旁组织和无淋巴结或远处转移大肠癌(26.5%、55.6%,P<0.05)。但此基因表达与大肠癌病理类型无显著相关性。点杂交和Northern-blot证实了RT-PCR结果。而胃癌与配对癌旁正常组织中未发现NGX6表达水平改变(表达下调率:29.4%vs41.2%,P>0.05).结论:NGX6基因在大肠癌中表达下调,此基因的低表达在大肠癌的发生和转移中起着一定的作用,但可能与胃癌的发生无关.AIM:To investigate the expression level of NGX6 in humangastric cancer and colorectal tissues cancer tissues as wellas their matched paracancerous tissues,and to investigatewhether this gene play a role in the pathogenesis of gastricand colorectal carcinomas.METHODS:The expression level of NGX6 in 34 cases ofhuman gastric cancer tissues,34 cases of human colorectalcancer tissues,and their matched paracancerous tissues wasanalyzed by RT-PCR,Dot hybridization and Northern blotanalysis.RESULTS: The down-regulation rate of NGX6 expressionin colorectal carcinomas tissues (73.5%,25/34)and the car-cinomas with lymph-node or distance metastasis ( 93.8%,15/16 ) was significantly higher than that in paracanceroustissues and the ones without metastasis (26.5% ,55.6%) (P<0.05), respectively .However the expression of NGX6 wasnot relevant to pathologic type of colorectal carcinomas.Dot and Northern hybridization analysis confirmed the re-sults of RT-PCR. The expression level of NGX6 did not dis-play any difference between gastric cancer and correspond-ing normal tissues.CONCLUSION: Expression of NGX6 gene downexpressedin colorectal carcinoma tissues,Low-expression of this genemay be associated with tumorgenesis and metastasis ofcolorectal carcinoma,but not with gastric carcinoma.
