摘 要:目的研究成纤维细胞生长因子(FGFR)抑制剂BGJ398对人源肾癌细胞株OSRC-2增殖、黏附及其对细胞侵袭功能的影响,为肾细胞癌的靶向治疗提供新思路。方法用不同浓度(0、0.1、0.5、1、2、5、10μmol/L)的FGFR抑制剂BGJ398处理体外培养的人源肾癌细胞OSRC-2后,通过CCK8法检测细胞增殖变化;激光共聚焦显微镜观察BGJ398对细胞的活力和骨架的影响;Transwell小室法检测BGJ398对人肾癌细胞侵袭能力的影响。结果 BGJ398能明显抑制OSRC-2细胞增殖。CCK8结果显示BGJ398处理人肾癌细胞后,细胞增殖受到抑制,并具有剂量依赖性。Live/Dead荧光染色实验证明BGJ398可显著抑制肾癌细胞的增殖及活力(P<0.01)。细胞黏附及Transwell实验结果表明,BGJ398可抑制细胞骨架的重塑及侵袭效应(P<0.01)。结论 BGJ398对肾癌细胞的增殖,活力和侵袭力具有明显的抑制作用。抑制FGFRs有望成为肾癌治疗的新靶点,为FGFR特异性抑制剂成为潜在的肾癌靶向治疗药物提供实验证据。Objective To evaluate inhibitory effects of fibroblast growth factor(FGFR) inhibitor BGJ398 on the proliferation, adhesion, and invasion of human renal cell carcinoma cells(OS-RC-2). To explore the new targeted therapy for renal cell carcinoma. Methods Human renal cell carcinoma cells OS-RC-2 were dealed with different concentration of 0, 0.1, 0.5, 1, 2, 5,10 μmol/L BGJ398. The survival of cells was supplied by CCK-8 assay. The viability and cytoskeleton of OS-RC-2 cells was checked by confocal fluorescence microscope. The invasion of OS-RC-2 cells was evaluated by transwell chamber methods. Results BGJ398 could inhibit the proliferation of OS-RC-2. CCK-8 assay revealed that BGJ398 could suppress the survival of OS-RC-2 by concentration dependant manner. Live/dead fluorescence staining experiments showed that BGJ398 can significantly inhibit the proliferation and viability of OS-RC-2 cells(P〈0.01). Cell adhesion and transwell chamber results show that BGJ398 could inhibit cytoskeleton remodeling and invasion of OS-RC-2 cells(P〈0.01). Conclusion BGJ398 can inhibit the proliferation, viability, and invasion of human renal cell carcinoma cells. Inhibition of FGFR could be the therapeutic targets of renal cell carcinoma, and provided experimental evidence for potential targeted therapies of FGFR inhibitors.
