摘  要:目的研究2-ME联合抗坏血酸在诱导白血病细胞凋亡中的作用以及相关作用机制。方法用显微镜观察K562细胞凋亡情况;通过流式细胞术分析凋亡率;通过EMSA检测NF-κB的活性。结果 2μmol/L2-ME+62.5μmol/LAA组可诱导K562细胞凋亡。2-ME+AA以剂量依赖性方式诱导凋亡,在浓度(2~8μmol/L)时凋亡呈时间-剂量依赖。2-ME可诱导NF-κBDNA结合活性以剂量依赖性方式降低。结论 2-ME联合抗坏血酸可诱导白血病细胞以时间-剂量依赖式凋亡,其诱导凋亡的机制与NF-κB通路有关。Objective To investigate the role of 2-methoxyestradiol combined with ascorbic acid in the apoptosis of leukemia K562 cells and its related molecular mechanisms. Methods The apoptosis of K562 cells was observed by a microscope. The apoptosis rate was analyzed by flow cytometry. The activity of NF-KB was detected by EMSA. Results The apoptosis of K562 cells was induced by 2 μmol/L 2-ME + 62.5 μmol/LAA. 2-ME + AA induced apoptosis in a dose- dependent manner. The apoptosis was time-dose dependent at the concentration (2-8 μmol / L). 2-ME could induce NF-KB DNA binding activity in a dose-dependent manner. Conclusion 2-methoxyestradiol combined with ascorbic acid can induce the apoptosis of leukemia K562 cells in a time-dose-dependent manner. The mechanism is related to the pathway of NF-KB.
