摘  要:【目的】探讨HOTAIR干扰质粒(siHOTAIR)对肝癌移植瘤裸鼠侵袭及转移相关信号通路的影响。【方法】将导入低表达长链非编码(long non-coding RNA,lnc RNA)Hotair质粒和空载质粒用重组慢病毒的人肝癌细胞系HepG2细胞分别接种裸鼠,通过实时荧光定量PCR及免疫组化分别检测肝癌移植瘤裸鼠侵袭及转移相关血管内皮生长因子(vascular endothelial growth factor,VEGF)/血管内皮生长因子受体(vascular endothelial growth factor receptor,VEGFR)m RNA和蛋白的表达情况。【结果】体外实验结果均显示Hep G2细胞组VEGF和VEGFR mRNA的相对表达量及蛋白的表达水平较si-HOTAIR Hep G2细胞组明显升高。【结论】si-Hotair通过靶向调控VEGF/VEGFR信号通路抑制肝癌的增殖发育及侵袭能力,未来有望成为肝癌临床治疗中新的着眼点。【Objective】To explore the effect of si-HOTAIR on the invasion-and-metastasis-related signaling pathway of transplanted tumors in nude mice with hepatocellular carcinoma. 【Methods】The nude mice were inoculated with Hep G2 cells which were transfected with the plasmid of low expression Lnc RNAHotair and empty vector into recombinant lentiviruses. The m RNA and protein expressions of VEGF/VEGFR related to the invasion and metastasis of transplanted tumors were detected by q RT-PCR and immunohistochemistry in nude mice with hepatocellular carcinoma.【Results】The results of in vitro experiments showed that the relative m RNA and protein expression levels of VEGF and VEGFR in Hep G2 cells were significantly higher than those in si-HOTAIR Hep G2 cells.【Conclusion】The proliferation and invasion of hepatocellular carcinoma are suppressed by si-HOTAIR via the targeted regulation of VEGF/VEGFR signaling pathway. It is expected to become a new focus in the clinical treatment for liver cancer in the future.
