摘  要:目的观察雌二醇对去卵巢大鼠焦虑样行为及杏仁核c-fos表达的影响,探讨雌二醇改善去卵巢大鼠焦虑样行为的可能机制。方法 30只SD雌性大鼠随机分为假手术组、卵巢切除组和去卵巢雌二醇组。雌二醇治疗8周后通过高架迷宫实验观察雌二醇对去卵巢大鼠焦虑样行为的影响;随后采用免疫组织化学结合图像半定量方法对杏仁核c-fos的表达进行测量和分析。结果与假手术组大鼠比较,高架迷宫实验中卵巢切除组大鼠开放臂次数及时间、开放臂次数比例及时间比例均显著下降(P<0.01),杏仁核c-fos数量及面积均显著增加(P<0.05);而去卵巢雌二醇组去卵巢大鼠的焦虑行为可显著改善,杏仁核c-fos表达减弱(P<0.05)。结论卵巢摘除会导致大鼠焦虑样行为,长期雌二醇治疗可改善去卵巢大鼠的焦虑行为,可能与雌二醇降低去卵巢大鼠杏仁核c-fos的表达有关。Objective To explore the influence of estradiol on anxiety-like behavior and amygdala c-fos expression and the possible mechanism of estradiol improving anxiety-like behavior of the ovariectomized( OVX) rats. Methods Thirty adult SD female rats were randomly assigned into 3 groups: sham group,OVX group and OVX + E2 group( ovariectomized rats treated with estradiol). The elevated plus maze was used to assess the anxiety-like behavior after 8-week estradiol treatment. Immunohistochemical staining was used to detect the c-fos expression in amygdala. Results Compared with sham group,open arm entry( OE),open arm time( OT),the percentage of OE and the percentage of OT significantly decreased in OVX group( P〈0. 01),while the number and area of amygdala c-fos expression significantly increased( P〈0. 05). Compared with OVX group,anxiety-like behavior was improved and amygdala c-fos expression decreased after 8-week estradiol treatment in OVX + E2group( P〈0. 05). Conclusion The ovariectomy may induce the anxiety-like behavior,and estradiol may ameliorate the anxiety-like behavior in OVX rats by decreasing the amygdala c-fos expression.
