摘 要:目的研究双链RNA-依赖的蛋白激酶(the double stranded RNA-dependent protein kinase,PKR)在阿尔茨海默病(Alzheimer’s disease,AD)患者外周血淋巴细胞(Peripheral lymphocytes,PBLs)中的表达及活性;探讨其水平及活性改变与AD患者认知障碍的相关性。方法选取AD、PD及健康老年人各20例,应用蛋白质免疫印迹技术检测PBLs中总PKR及磷酸化PKR(p-PKR)的表达,分析比较3组中PKR表达及活性的差异,并利用皮尔森相关分析探讨PKR表达及活性与AD、PD病程和病情的关系。结果与正常对照组相比,总PKR、p-PKR蛋白表达水平在AD患者的PBLs中均明显升高(P<0.05),且总PKR的表达量与AD患者认知障碍严重程度呈正相关(r=0.799,P<0.01);在PD组PBLs中总的PKR与p-PKR蛋白水平均升高(P<0.05)。结论在AD患者中,PKR的异常表达及活化不仅体现在中枢神经系统,而且可在PBLs中检测到,其表达量与认知障碍严重程度成正相关,有可能成为AD早期诊断及病情评价的外周标记物。Objective To investigate PKR and its activated status levels of expression in peripheral lymphocytes( PBLs) of Alzheimer's disease( AD),and to discuss the relationship of this change and cognitive disfunction. Methods Healthy control,AD and PD were set up,containing 20 cases respectively. PKR and phospholated PKR( p-PKR) levels of expression in lymphocytes were detected by Western blotting and the changes of total and activated PKR levels were analyzed in three groups. The relationships of PKR and p-PKR expression levels and diseases durations and severities of AD and PD were discussed by Pearson's correlation analysis. Results Both total PKR and activated status levels were augmented in PBLs of AD patients compared with controls( P〈0. 05) and total PKR and p-PKR levels were positively correlated with the cognitive severity( r = 0. 799,P 〈0. 01). Both PKR and p-PKR levels were increased in lymphocytes of PD patients compared with controls( P〈0. 05). Conclusion The abnormal expressions of PKR and p-PKR were not only in central neurological system but also in PBLs. Total PKR and p-PKR levels were positively correlated with the cognitive severity of AD patients. Thus PKR may be a potential peripheral biomarker in early diagnosis and cognitive function estimations during AD processes.
