摘 要:目的:制备PLGA-壳聚糖/大黄素电纺纤维膜,并评价其相关理化性能。方法:分别采用同轴静电纺丝法和传统静电纺丝法(混纺法)制备PLGA-壳聚糖/大黄素电纺纤维膜;扫描电镜观察纤维膜的表面形貌,透射电镜观察纤维的同轴结构,生物力学测试仪测量纤维膜力学的性能,高压液相色谱(HPLC)法测量大黄素的释放率。结果:利用同轴静电纺丝技术制备的同轴PLGA-壳聚糖/大黄素电纺纤维膜的纤维方向随机且直径均匀,纤维表面光滑无串珠;其纤维直径为(927±73)nm,拉伸强度为(4.62±0.58)MPa,弹性模量为(13.73±2.64)MPa,均明显高于混纺组(P<0.05),而大黄素的释放则较混纺组明显变缓(P<0.05),并可持续释放15 d以上。结论:同轴静电纺丝法制备的PLGA-壳聚糖/大黄素电纺纤维膜具有良好的力学性能和药物缓释效果。AIM To prepare a novel Polylactide ( PLA) - chitosan( CS)/emodin nanofibrous membrane and to evaluate the physical and chemical properties of PLA - CS/Emodin. METHODS : Nanofibrous membrane with PLA - CS/Emodin was preparedb by co - electrospinning technology (test group) and traditional electrospinning meth-od (the control group) respectively. Then morphology and coaxial structure was observed by SEM, the mechanical properties and emodin release rate were examined by biomechanic tester and HPLC respectively. RESULTS : The fiber of the test group was uniformly distributed and the surface of fibers was smooth. The diameter was (927 ±73) nm, and the tensile strength (4. 62 ± 0. 58) MPa,the elastic modulus( 13. 73 ± 2. 64) MPa,emodin release rate was lower than that of the control ( P 〈 0. 05 ) and the release kept for more than 15 d. The mechanical properties of the test group were different from those of the control group. CONCLUSION : The PLA - CS/Emodin fiber presents outstanding mechanical properties and sustained - release properties.
