摘  要:通过响应面法确定不同年份黄精中总糖含量的最佳测定条件,在单因素试验的基础上,选定浓硫酸质量分数、蒽酮用量和显色时间3个因素,利用响应面分析法考察各因素对吸光度的影响,结合3,5-二硝基水杨酸法对黄精中还原糖含量进行测定,并比较不同年份中多糖含量。结果表明:最优测定条件为蒽酮用量0.2 g,浓硫酸质量分数为87%,显色时间为25 min;经过测定,4年生黄精总糖和还原糖含量均高。该方法适合黄精总糖含量的测定,可用于评价黄精药材的质量和为黄精的采收时间提供理论依据。This study aimed to confirm the optimal conditions of total sugar content in Polygonatum sibMcum Red. of different years with response surface methodology. In the anthrone-sulfuric acid method, three experimental conditions including concentrated sulfuric acid mass fraction, anthrone dosage and color development time were optimized by single factor experiments and response surface methodology using absorbance value as the dependent variable. The method combined with 3, 5-dinitrosalicylic acid method to determine the reducing sugar content of Polygonatum sibirieum Red. and compared the polysaccharide content in different years. Result showed that the optimum experimental conditions were found as follows: 87% concentrated sulfuric acid, 0.2 g of anthrone dosage and 25 min of color development. The total sugar and reducing sugar content ofPolygonatum sibiricum Red. of four-year were the highest in test. The method was suitable for the determination of Polygonatum sibiricum Red. total sugar, which could be used for quality evaluation of Polygonatum sibiricum Red. and its harvesting time.
