摘  要:目的探讨哺乳期小窝喂养对大鼠肥胖及脂肪酸合成酶(FAS)、腺苷酸活化蛋白激酶α(AMPKα)及磷酸化AMPKα(pAMPKα)表达的影响,探索早期防治肥胖的靶标和措施。方法新生雄性Wistar大鼠随机分为对照组(8只/乳母)和小窝组(5只/乳母),3w断乳后均给予标准膳食喂养至成年(第8w末),对照组大鼠按体重随机分为两组:对照组(仍食用标准膳食)和高脂对照组。高脂对照组和小窝组大鼠均高脂膳食诱导肥胖至14w末。分期处死大鼠取材,计算体脂含量,酶法检测血脂,FQ-PCR法检测AMPK、FAS基因的表达,Western blot法检测AMPKα和pAMPKα蛋白的表达。结果与对照组相比,在3、8和14w末,小窝组的体重、体脂含量和血清甘油三酯(TG)均升高(P<0.05);AMPK基因和蛋白的表达持续降低,FAS基因的表达持续升高(P<0.05)。结论哺乳期小窝喂养可增加血清TG、体脂含量和成年肥胖发生的风险;其机制可能与持续影响AMPK/FAS信号通路基因或蛋白的表达相关。Objective To evaluate the effects of small litters of rat pups during lactation on adult body weight, mRNA or protein expression of AMPK and FAS, and to find the measures and pharmacological targets of early prevention and treatment of obesity. Methods Newborn male Wistar rats were randomly divided into control group (8 pups/dam) and small litters group (5 pups/dam). In the adult period,the rats were fed with a high - fat diet to induce obesity. Serum triglyceride was assayed using the standard biochemical technique, mRNA or protein levels of AMPK and FAS were measured by quantitative PCR and western blotting. Results Compared with the corresponding controls at 3,8 and 14 weeks of age, body weight, fat content and serum TG increased in the small litters group accompanied by down - regulated mRNA and protein expression of AMPK and up - regulated mRNA expression of FAS. Conclusion The results suggest that the small litters during sucking increased later - life obesity, which might be related to programming of the expression of AMPK signal pathways.
