摘 要:非髓样甲状腺癌(nonmedullary thyroid cancer,NMTC)是指甲状腺滤泡上皮细胞起源的甲状腺癌,其发病率高居不下,一直是流行病学研究的热点之一。越来越多的研究表明,遗传因素在NMTC发生中占据重要位置。为寻找NMTC的易感基因,多年来对多个候选基因与NMTC的关联性进行了分析,由于多种原因没有成功发现明确的NMTC的易感基因。近年来全基因组关联分析、全外显子测序分析等技术开始运用于NMTC遗传学研究,得到了一些有意义的初步结果,但需进一步明确候选基因与通路在NMTC发生中的作用。这些分析指出稀有变异,而不是常见变异,对NMTC发生起了主要贡献。文章对NMTC的遗传学研究最新进展做了综述。Nonmedullary thyroid cancer (NMTC) is defned as the thyroidfollicular epithelial cells-derived tumour. NMTC is rapidly increasingin incidence. A large mount of studies have shown that genetics play animportant role in the pathogenesis of NMTC. For finding susceptibilitygenes, the associations between many candidate genes and NMTC havebeen explored. However, no susceptibility gene has been identifed becauseof the complexity and heterogeneous of NMTC. Recently, advancedgenetic and genomic technology (genomic-wide association and whole-exome sequencing) have been introduced into the studying the genetics ofNMTC, which requiring further investigation. All genetic studies suggestthat it might be rare variants, but not common variants, contribute thepathogenesis of NMTC. This review will briefy discuss the update progressin the predisposition of nonmedullary thyroid cancer.
