摘 要:目的探讨水通道蛋白4(AQP4)在肺癌裸小鼠模型中的表达及其对肿瘤细胞增殖与迁移的影响。方法2016年12月于昆明医科大学第三附属医院云南省肺癌重点实验室进行实验,选取SPF级雄性裸小鼠60只并采用随机数字法分为对照组与观察组,每组30只。对照组裸小鼠于背侧靠腋窝处皮下注射未转染的A549肺癌细胞0.2 ml以建立肺癌裸小鼠模型,而观察组裸小鼠于背侧靠腋窝处皮下注射AQP4-shRNA转染的A549肺癌细胞0.2 ml以建立肺癌裸小鼠模型。比较两组裸小鼠肿瘤重量、微血管密度(MVD),AQP4、E-钙黏着蛋白(E-cadherin)、N-钙黏着蛋白(N-cadherin)、波形蛋白(Vimentin)表达水平。结果观察组裸小鼠肿瘤重量及MVD低于对照组(P<0.05)。观察组裸小鼠AQP4、N-cadherin、Vimentin表达水平低于对照组,E-cadherin表达水平高于对照组(P<0.05)。结论APQ4在肺癌裸小鼠模型中表达增强,其可能参与了肺癌的病理生理过程,可在一定程度上促进肿瘤细胞的增殖与迁移。Objective To investigate the expression and impact on tumor cell proliferation and migration of AQP4 in nude mice with lung cancer. Methods This experiment was carried out in December 2016, in Yunnan Provincial Key Laboratory for Lung Cancer,the Third Affiliated Hospital of Kunming Medical University. A total of 60 SPF-level male nude mice were selected and divided into control group and observation group according to random number table, each of 30 mice. Nude mice of control group received subcutaneous injection of A549 lung cancer cells without infection( 0. 2 ml) around dorsal axillary tissue to prepare the lung cancer model,while nude mice of observation group received subcutaneous injection of A549 lung cancer cells with AQP4-shRNA infection( 0. 2 ml) around dorsal axillary tissue to prepare the lung cancer model. Tumor weight,microvessel density,expressions of AQP4,E-cadherin,N-cadherin and Vimentin were compared between the two groups. Results Tumor weight and microvessel density of observation group were statistically significantly lower than those of control group( P〈0. 05). Expressions of AQP4, N-cadherin and Vimentin of observation group were statistically significantly lower than those of control group,while expression of E-cadherin of observation group was statistically significantly higher than that of control group( P〈0. 05). Conclusion Expression of APQ4 is higher in nude mice with lung cancer,it may plan a role in the pathophysiological process of lung cancer,can promote the tumor cell proliferation and migration to some extent.
