摘 要:随着生物技术的发展,近年来可用于临床的蛋白多肽类药物日益增多,且已成为治疗众多疾病的首选药物。蛋白多肽类药物在药代动力学方面仍有许多问题需要解决,该类药物的吸收、分布、代谢、排泄过程与小分子药物不同,各种内源性蛋白的干扰也对该类药物的分析造成影响。综述蛋白多肽类药物药代动力学分析方法研究进展,重点介绍同位素标记示踪法、活体成像技术、免疫分析法、色谱法等方法的特点及其在蛋白质多肽类药物中的应用。With the development of biotechnology, an increasing number of protein and peptide drugs have been available for clinical settings in recent years and become the first choice of therapy for many diseases. However, in the aspect of pharmacokinetics, many fundamental problems need to be addressed. The underlying mechanisms for the processes of absorption, distribution, metabolism and excretion (ADME) of protein and peptide drugs are very different from that of small molecule drugs. Various endogenous proteins will also interfere with quantitative analysis. This review provides recent progress in pharmacokinetics of protein and peptide drugs with emphasis on the characteristics of analytic methods like radioiodination, #1 vivo imaging techniques, immunoassay, and chromatography as well as their applications in the study of protein and peptide drugs.
