摘  要:目的:研究还原型谷胱甘肽是否可以影响伊立替康导致的肝脏脂肪变性的发生。方法:建立结肠癌HCT116细胞裸鼠皮下移植瘤模型,皮下注射伊立替康单药或联合还原型谷胱甘肽,7天后处死裸鼠,取肝脏制作肝脏病理切片观察肝脏损伤情况并制备10%肝脏组织匀浆检测肝脏组织内丙二醛(MDA)及超氧化物歧化酶(SOD)的表达情况。结果:成功建立了结肠癌HCT116细胞裸鼠皮下移植瘤模型;联合还原型谷胱甘肽组肝脏脂肪变性的发生率降低,肝脏SOD的表达较不联合还原型谷胱甘肽升高(P<0.05);MDA的表达较不联合还原型谷胱甘肽降低(P<0.05)。结论:还原型谷胱甘肽可以通过增强肝细胞的清除氧自由基能力抑制脂质过氧化过程,最终起到保护肝脏的作用,降低伊立替康引起的肝脏脂肪变性的发生率。Objective:To evaluate if glutathione can reduce the incidence rate of steatohepatitis caused by irinote- can. Methods:Nude mice sub skin model was constructed by transplanting human colocarcinoma HCTll6 cells, then injected irinotecan with or without reduced -glutathione. 7 days later, nude mice were sacrificed. The hepatic in- jury was observed by pathological section and HE staining and 10 percent of the liver homogenate was prepared to de- tect the superoxide dismutase (SOD) and malondialdehyde (MDA) activity. Results: Nude mice sub - skin model transplanted human colocarcinoma HCTll6 ceils was constructed. The incidence rate of steatohepatitis was lower in treatment group than the control group. The expression of SOD was higher in treatment group than the control group and MDA was lower. Conclusion:Nude mice sub - skin model transplanted human colocarcinoma HCT116 cells was constructed. The incidence rate of steatohepatitis was lower in treatment group than the control group. The expression of SOD was higher in treatment group than the control group and MDA was lower.
