摘 要:目的通过检测健康志愿者、肝硬化患者和原发性肝癌患者血清中粒细胞—巨噬细胞集落刺激因子(GM-CSF)、核苷酸结合寡聚化结构域样受体3(NLRP3)和Toll样受体4(TLR4)的含量,探讨GM-CSF、NLRP3和TLR4在原发性肝癌疾病过程中的可能作用。方法收集肝癌手术切除的50例肝癌组织及癌旁组织标本,以及肝硬化和肝癌患者血清标本各12例,健康志愿者12例。采用酶联免疫吸附试验(ELISA)和实时荧光定量PCR方法分别检测血清中mRNA水平上GM-CSF、NLRP3和TLR4的表达。结果正常肝细胞GM-CSF、NLRP3和TLR4 mRNA的表达水平均显著高于肝癌细胞(P<0.001);原发性肝癌组织GM-CSF、NLRP3和TLR4 mRNA的表达水平均明显低于癌旁组织(P<0.001);健康志愿者血清GM-CSF、NLRP3和TLR4的含量显著高于肝硬化及肝癌患者(P<0.001),但肝硬化患者和肝癌患者血清GM-CSF、NLRP3和TLR4的含量差异无统计学意义(P>0.05)。结论肝癌组织及癌细胞系GM-CSF、NLRP3和TLR4与癌旁组织及人正常肝细胞相比表达显著降低,GM-CSF、NLRP3和TLR4信号途径可能参与了原发性肝癌的免疫过程。Objective This study detecting the expression of granulocyte-macrophage colony stimulating factor (GM-CSF), nod-like receptor protein 3 (NLRP3) and toll-like receptor 4 (TLR4) in healthy volunteers and the patients of liver cirrhosis and hepatic celluler cancer, explore the possible role of intestinal immune factor GM- CSF, NLRP3 and TLR4 in the process of primary liver cancer disease. Methods The expression of GM-CSF, NLRP3 and TLR4 was detected in serum by ELISA and in mRNA level by RT-PCR. Results In the mRNA level, the expression of GM-CSF, NLRP3 and TLR4 of carcinoma tissue were significantly lower than tissue adjacent to carcinoma (P 〈 0.001); The expression of GM-CSF, NLRP3 and TLR4 of normal liver cells were significantly higher than that of liver cancer cells (P 〈 0.001); ELISA showed that the expression of GM-CSF, NLRP3 and TLR4 in healthy volunteers were significantly higher than that of patients with liver cirrhosis and liver cancer, but no significant difference between cirrhosis and liver cancer group. Conclusion The expres- sion of GM-CSF, NLRP3 and TLR4 in liver cancer tissue and cancer cells were relatively suppressed, com- pared with tissue adjacent to carcinoma and normal liver cells. GM-CSF, NLRP3 and TLR4 signaling path- ways may be involved in the immune process of liver cancer.
