摘  要:目的:研究肌球蛋白轻链(MYL9)在非小细胞肺癌(NSCLC)患者的淋巴结和外周血中的表达及其与临床病理特征的关系。方法:选取在2015年4月至2016年10月广西医科大学第一附属医院胸外科,并行肺叶切除+系统性淋巴结清扫手术治疗的60例NSCLC患者作为研究组,同期选取60例无恶性肿瘤病史的健康体检者为对照组。采用实时荧光定量PCR(q PCR)及western blotting法检测MYL9在淋巴结和外周血中m RNA和蛋白表达水平;多因素Logistic回归模型分析MYL9m RNA及蛋白表达的影响因素。结果:研究组外周血MYL9 m RNA及蛋白表达量均高于对照组(均P<0.05);MYL9 m RNA和蛋白在淋巴结转移阳性患者的表达量均高于淋巴结转移阴性患者的表达量,差异有统计学意义(均P<0.05);经多因素Logistic回归分析,肿瘤分化程度和淋巴结分期是影响NSCLC患者MYL9表达的独立危险因素(P<0.05)。结论:NSCLC患者肿瘤分化程度越差,淋巴结分期越晚,MYL9 m RNA和蛋白表达上调越明显;MYL9可能参与了NSCLC的转移过程,是一个潜在的预后预测指标。Objective: To investigate the expression of myosin light chain 9 (MYL9) in lymph nodes and periph- eral blood in patients with non-small cell lung cancer (NSCLC), and to explore the relationship between MYL9 ex- pression and the clinicopathologic features. Methods: 60 patients with NSCLC who underwent pulmonary lobecto- my and systematic lymph node dissection in our hospital from April 2015 to October 2016 were selected as a study group. 60 healthy subjects who had no history of cancer were enrolled as a control group. The mRNA and protein expressions of MYL9 in the lymph nodes and peripheral blood were detected by real-time fluorescence quantitative PCR (qPCR) and western blotting, respectively, and the influencing factors were analyzed by multivariate logistic regression analysis. Results: MYL9 mRNA and protein expressions in peripheral blood were higher in the study group than those in the control group (P〈0. 05). Patients with lymphatic metastasis had higher level of MYL9 ex- pression (P〈0. 05). Multivariate logistic regression analysis revealed that tumor differentiation and lymph node staging were independent risk factors for MYL9 expression in patients with NSCLC. Conclusion: MYL9 might be involved in the process of tumor metastasis in NSCLC, and was a potential prognostic indicator.
