摘 要:目的:探讨川芎提取液对D-半乳糖致衰老小鼠学习记忆的影响及其与血清、脑、肝脏、肾脏等组织中SOD活力、GSH-Px活性和MDA含量的关系。方法:采用小鼠颈背部皮下连续注射D-半乳糖90d建立衰老小鼠模型,衰老模型成功的小鼠分为3组:衰老模型组、生理盐水对照组、川芎提取液组,每组8只,川芎提取液组按12g/kg连续灌胃14d,生理盐水对照组灌与川芎治疗组相同容量的生理盐水,衰老模型组小鼠不灌任何东西。另8只小鼠不注射D-半乳糖作为正常对照组。Y型迷宫实验测试小鼠学习记忆能力;对小鼠血清、脑、肝脏、肾脏采样,测定其中超氧化物歧化酶(SOD)活力、谷胱甘肽过氧化物酶(GSH-Px)活性和丙二醛(MDA)含量。结果:与正常对照组相比,衰老模型组学习测试次数和记忆错误次数均明显增多;与衰老模型组相比,川芎提取液组学习测试次数和记忆错误次数均明显减少,差异均有统计学意义(P<0.05)。与正常对照组相比,衰老模型组血清、脑、肝、肾脏SOD活力和GSH-Px活性均明显降低而MDA含量均明显升高;与衰老模型组相比,川芎提取液组血清、脑、肝、肾脏SOD活力和GSH-Px活性均明显升高;而MDA含量均明显降低,差异均有统计学意义(P<0.05)。结论:川芎可能通过升高血清、脑、肝、肾脏SOD活力和GSH-Px活性,降低MDA含量,从而改善衰老小鼠学习记忆能力。Objective:To explore the effect of Ligusticum chuanxiong hort abstract to the ability ot learning and memory and the levels of SOD, GSH-Px and MDA in serum, brain, liver and kidney in aging mice treated with D-galactose. Methods: Made aging mice with D-galactose for 90 days. Divided aging mice into 3 groups: aging mice model group, saline group and Ligustieum chuanxiong hort abstract group. Each group had 8 mice. Ligusticum chuanxiong hort abstract group was treated Ligusticum chuanxiong hort abstract 12g/kg for 14 days. The control group was clear with Dgalactose. The spatial learning and memory ability were evaluated by Y-maze. Mice's serum, brain, liver and kidney were collected and the activity of SOD and GSH-Px, the level of MDA were measured. Results:Compared with the con- trol group, the learning ability of aging mice group were significantly increased (P〈0. 05) ,the memory ability of Ligusticum chuanxiong hort abstract group was significantly increased (P〈0. 05) ;the activity of SOD, GSH-Px in serum, brain, liver and kidney decreased significantly in aging mice group compared with the control roup(P〈0. 05) ; the activity of MDA in serum, liver,kidney and heart increased significantly in aging mice group compared with the control group(P〈0. 05). the activity of SOD, GSH-Px in serum, brain, liver and kidney increased significantly in Ligusticum chuanxiong hort abstract group compared with the control group(P〈0.05) ;the activity of MDA in serum, brain, liver and kidney decreased significantly in aging mice group compared with the control group(P〈0. 05). Conclusion: Ligusticum chuanxiong hort increases the activity of SOD and GSH-Px, decreases the level of MDA in mice' s serum,brain, liver and kidney and improves the ability of learning and memory.
