摘 要:目的:探讨S100A6、膜联蛋白A2(annexin A2,AnxA2)及c-myc在多发性骨髓瘤(multiple myeloma,MM)患者中的表达及其临床意义。方法:应用实时荧光定量PCR法检测28例MM患者化疗前后及20例对照组(外周血白细胞或血小板计数略低于正常,骨髓检查正常者)骨髓单个核细胞中S100A6、AnxA2及c-myc mRNA的表达,分析MM患者S100A6、AnxA2及c-myc mRNA表达间的关系。将S100A6 si RNA转染至MM U266细胞后,分别应用实时荧光定量PCR及蛋白质印迹法检测S100A6、AnxA2及c-myc mRNA和蛋白的表达。结果:MM患者骨髓单个核细胞中S100A6、AnxA2及c-myc mRNA的表达水平均高于对照组(P值均<0.05),化疗前MM患者骨髓单个核细胞中S100A6、AnxA2及c-myc mRNA的表达水平均高于化疗后(P值均<0.05),伴髓外转移的MM患者骨髓单个核细胞中S100A6、AnxA2及c-myc mRNA的表达水平均高于不伴髓外转移的患者(P值均<0.05),并且S100A6 mRNA的表达水平与AnxA2 mRNA、c-myc mRNA表达水平均呈正相关(r=0.585,P=0.001;r=0.540,P=0.004)。S100A6 si RNA转染后,MM U266细胞中S100A6、AnxA2及c-myc mRNA和蛋白的表达水平均下调(P值均<0.05)。结论:S100A6在MM骨髓中的表达水平较高,且与AnxA2和c-myc的表达呈正相关。S100A6可能与MM的发生、进展和髓外转移相关。Objective: To investigate the expressions of S100A6, annexin A2(AnxA2) and c-myc in patients with multiple myeloma(MM) and their clinical significance.Methods: The expressions of S100A6, AnxA2 and c-myc mRNA sin bone marrow mononuclear cells from 28 cases of MM before and after chemotherapy and 20 controls(wh o s e peripheral blood white cell count and the platelet count were a little lower than the normal values, but the result of bone marrow aspiration was normal) were detected by real-time fluorescent quantitative PCR. The relationships among the expressions of S100A6, AnxA2 and c-myc m R N A s were analyzed. The expressions of S100A6, AnxA2 and c-myc mRNAs and proteins in MMU266 cells after transfection with S100A6 si RNA were detected by real-time fluorescent quantitative PCR and Western blotting, respectively. Results: The expression levels of S100A6, AnxA2 and c-myc mRNAs in bone marrow mononuclear cells from patients with MM were higher than those from the controls(all P〈 0.05). The expression levels of S100A6, AnxA2 and c-myc mRNAs in bone marrow mononuclear cells from patients with MM before chemotherapy were higher than those after chemotherapy(all P〈 0.05). The expression levels of S100A6, AnxA2 and c-myc mRNA s i n bone marrow mononuclear cells from MM patients with extramedullary metastasis were higher than those from MM patients not having extramedullary metastasis(all P〈 0.05). The expression of S100A6 m R N A was positively correlated with the expressions of AnxA2 and c-myc mRNAs(r = 0.585, P = 0.001; r = 0.540, P =0.004). The expression levels of S100A6, AnxA2 and c-myc mRNAs and proteins in MMU266 cells after transfection with S100A6 si RNA were down-regulated(all P 〈0.05).Conclusion: The expression level of S100A6 in patients with MM is higher, and it is positively associated with AnxA2 and c-myc. S100A6 may be involved in the development, progression and extramedullary metastasis of MM.
