摘  要:目的基于响应面法优化水飞蓟油中油酸甲酯化率的工艺。方法在单因素试验的基础上,采用响应面分析法,以油酸甲酯化率为检测指标,以氢氧化钾用量、甲醇用量、反应时间为自变量进行甲酯化过程优化试验。结果通过响应面分析法后得到的最佳工艺为氢氧化钾用量为0.6 mol·L^(-1),甲醇用量为12 mL,反应时间为70 min,油酸甲酯化率可达到95.12%,在此条件下对水飞蓟油进行甲酯化反应,测得样品中油酸含量为23.52%,亚油酸含量为54.45%。结论采用响应面法对水飞蓟油中油酸甲酯化率进行优化合理可行,采用优化后的最佳工艺进行含量测定,对油酸的准确定量定性分析具有重要的意义。Objective To optimize the rate of methyl esterification in Silybum marianum seed oil by the response surface method. Methods On the basis of single factor experiment, the methyl oleate rate was used as the test index, and the dosage of potassium hydroxide, the dosage of methanol, and the reaction time were used as the independent variables in the optimization. Results The optimal addition was potassium hydroxide(0.6 mol·L^(-1)), methanol(12 mL) and reaction time(70 min). The rate of the methyl oleate reached 95.12% under the optimum condition. The content of oleic acid was 23.52% and the content of linoleic acid was 54.45% in the sample. Conclusion Response surface methodology is feasible in optimizing the rate of methyl esterification in Silybum marianum seed oil. The best optimized technology for content determination is important for the accurate quantitative analysis of oleic acid.
