摘 要:目的探讨青海地区藏、汉民族子痫前期患者与HLA-G基因Exon8 14bp插入/缺失多态性的相关性。方法收集2015年4月到2016年10月间在青海大学附属医院产科藏、汉民族正常及子痫前期孕妇血标本348例,采用DNA提取、聚合酶联反应、琼脂糖电泳及测序等方法对四组HLA-G的14 bp位点的等位基因及基因型进行分型,比较各组间该位点的等位基因及基因型分布是否存在差异。结果各组间等位基因及基因型多态性分布差异均无统计学意义,而将基因型合并后发现该位点在汉族正常组及藏族正常组间分布存在差异,差异具有统计学意义。结论藏、汉族患者HLA-G基因Exon8 14 bp插入/缺失的多态性可能与子痫前期的发病无关,而与民族差异有关。Objective To study the relationship between Preeclampsia(PE) and polymorphism of HLA-G exon8 14 bp insertion/deletion in Han and Tibetan nationality of Qinghai province.Methods Collect 348 blood samples of normal and PE pregnancies, of which delivered at Affiliated Hospital of Qinghai Univernity.Detect allele and genotype of HLA-G Exon 8 14 bp insertion/deletion by DNA extraction, polymerise chain reaction, agarose electro- ptohoresis and sequencing among the four groups, and compare whether there is difference in the distributions of al- lele and genotype located at the site of gene among the groups.Results There was no significant difference among the four groups in distributions of allele and genotype, however, combine with genotypes we found that there was a statistical significance in control group of Han and Tibetan nationality. Conclusion The polymorphism of the HLA-G gene Exon8 14 bp insertion / deletion in Tibetan and Han patients may not be related to the incidence of preeclampsia and is related to ethnic differences.
