摘 要:目的:检测口腔鳞状细胞癌组织中X盒结合蛋白1(XBP1)的表达及观察抑制其表达后对人口腔鳞状细胞癌Tca8113增殖凋亡的影响,并探讨其机制。方法:RT-PCR检测42例口腔鳞状细胞癌及癌旁组织中XBP1基因的mRNA表达;NC-siRNA和XBP1-siRNA转染至Tca8113细胞内,不经任何处理的细胞作为空白对照组,48h后,CCK8实验、流式细胞仪分别检测XBP1基因转染对细胞增殖、凋亡的影响;Western blot检测Cleaved caspase3、β-catenin、Cyclin D1蛋白表达的变化。结果:口腔鳞状细胞癌中XBP1基因的mRNA表达显著高于癌旁组织(P<0.01);转染XBP1-siRNA能够显著降低XBP1的蛋白表达,降低细胞的存活率,促进细胞的凋亡,上调Cleaved caspase3蛋白表达,下调β-catenin、Cyclin D1蛋白表达(P<0.01)。结论:RNA干扰沉默XBP1基因表达可通过抑制Wnt/β-catenin信号通路降低Tca8113增殖及诱导细胞凋亡。Objective: To detect the expression of X box binding protein 1 (XBP1) in human oral squamous cell carcinoma (OSCC) and to investigate the effect of inhibition the expression of XBP1 on proliferation and apoptosis of Tca8113. Methods: The expressions of XBP1 gene in the OSCC and paracancerous tissue were detected by RT-PC. NC-siRNA and XBP1-siRNA were transfected into Tca8113 cells, and the cells without any treatment were set the control group. The cell proliferation and apoptosis were detected after XBP1 gene transfection for 48h by CCK8 test and flow cytometry. The cleaved caspase3, β-catenin, and Cyclin D1 protein expression were detected by Western blot. Results: The mRNA expression of XBP1 gene in OSCC was significantly higher than that in paracancerous tissues (P〈0.01). The expression of XBP1 protein and the cell viability were significantly decreased. Cell apoptosis were promoted. The protein expression of cleaved Caspase3 was increased, but the expressions of β-catenin and Cyclin D1 were decreased after XBP1-siRNA transfection (P〈0.01). Conclusion: Silencing XBP1 gene expression by RNA interference can inhibit the proliferation of Tca8113 and induce apoptosis by inhibiting the Wnt/ β-catenin signaling pathway.
