摘  要:目的检测钴铬合金、银钯合金及纯钛金属全冠修复后犬牙龈细胞中Bcl-2和Bax基因m RNA的表达,探讨3种材料全冠修复对细胞凋亡的影响。方法选取5只成年雄性实验犬,将每只犬的牙弓分为4个区,以每区的尖牙为实验对象,分别行钴铬合金(A组)、银钯合金(B组)及纯钛(C组)金属全冠修复,第4区为空白对照(D组);分别于修复后1、2、3个月采集尖牙牙龈组织,用逆转录聚合酶链式反应(RT-PCR)法检测牙龈细胞中Bcl-2、Bax基因m RNA的表达并计算Bcl-2/Bax的比率。结果修复后3个月,A组Bcl-2/Bax的比率高于D组,差异有统计学意义(P<0.05);修复后2、3个月,B组Bcl-2/Bax的比率低于D组,差异有统计学意义(P<0.05);修复后1、2、3个月,C组Bcl-2/Bax的比率与D组相比,差异均无统计学意义(P>0.05)。结论钴铬合金抑制牙龈细胞凋亡,银钯合金促进牙龈细胞凋亡,而纯钛对牙龈细胞的凋亡无明显影响。Objective To observe the expression of Bcl-2 and Bax gene in gingival cells of the dogs' after restored with cobalt-chrome alloy full crown and palladium-silver alloy full crown and pure titanium full crown. Investigate the effects of three materials on apoptosis of the gingival cells. Methods 5 adult male dogs were chosen for the experiment, each dog's dental arch was divided into 4 sections. Each of the fang of each area was restored with cobalt-chrome alloy crown, palladium-silver alloy crown and pure titanium crown respectively, the fourth area is for control group. To collect the gingival tissue of fangs in 1, 2 and 3 months after restoration. The level of Bcl-2 and Bax gene expression were detected by RT-PCR. Results After 3 months restoration, the group A was superior to group D on the Bcl-2/Bax ratio with statistically significant difference(P〈0.05). After 2 and 3 months restoration, the group B was inferior to group D on the Bcl- 2/Bax ratio with statistically significant difference (P〈0.05). After l, 2 and 3 months restoration, the group C had no statistically significant difference with group D on the Bcl-2/Bax ration (P〉0.05). Conclusion The cobalt- chromium alloy inhibits the apoptosis of gingival cells, the palladium-silver alloy promotes the apoptosis of gingival cells, while the pure titanium has no obvious effect on the apoptosis of gingival cells.
