敲低KHDRBS3基因抑制人卵巢癌CAOV-3细胞增殖

摘　　要：目的探讨含KH RNA结合结构域信号转导相关分子3(KHDRBS3)基因对体外培养的人卵巢癌CAOV-3细胞增殖的影响。方法取对数生长期细胞,分为未转染组、无关小干涉RNA(siRNA)序列转染组、KHDRBS3-siRNA转染组。采用反转录PCR及Western blot法验证KHDRBS3基因的沉默效果;采用四甲基偶氮唑蓝(MTT)比色法比较不同组别CAOV-3细胞增殖情况;采用流式细胞术检测不同处理组CAOV-3细胞的细胞周期分布。结果 KHDRBS3-siRNA转染后可明显降低CAOV-3细胞KHDRBS3的mRNA和蛋白水平,敲低CAOV-3细胞KHDRBS3水平后,细胞的增殖能力明显降低,G0/G1期细胞比例增加、S期细胞比例降低、G2/M期细胞比例无明显变化。结论敲低KHDRBS3的水平可使CAOV-3细胞发生G0/G1期阻滞,抑制细胞增殖。Objective To investigate the effect of KH RNA binding domain-containing,signal transduction-associated 3（ KHDRBS3） gene on the proliferation of human ovarian cancer CAOV-3 cells in vitro. Methods The logarithmic growth phase cells were divided into non-transfection group,unrelated small interfering RNA（ siRNA） sequence transfection group and KHDRBS3-siRNA transfection group. Reverse transcription PCR and Western blotting were used to verify the result of KHDRBS3 gene silencing. The proliferation of CAOV-3 cells in different groups was compared by MTT assay. The cell cycle distribution of CAOV-3 cells in different treatment groups was detected by flow cytometry. Results KHDRBS3-siRNA transfection significantly decreased the mRNA and protein levels of KHDRBS3 gene in CAOV-3 cells. After the knock-down of KHDRBS3 gene in CAOV-3 cells,the proliferation of the cells markedly decreased,the percentage of G0/G1 phase cells increased,and the percentage of S phase cells decreased,while the percentage of G2/M phase cells did not change significantly. Conclusion Down-regulating the KHDRBS3 gene can cause G0/G1 phase arrest and inhibit cell proliferation in CAOV-3 cells.
