摘 要:目的探讨长链非编码RNA-linc00152对口腔鳞状细胞癌增殖和侵袭能力的影响。方法采用RT-q PCR方法检测linc00152在口腔鳞状细胞癌组织、癌旁组织以及口腔鳞癌细胞系中的表达。CAL27和SCC9细胞转染linc00152-siRNA后,采用CCK8、划痕以及Transwell实验检测CAL27和SCC9细胞增殖、侵袭转移情况。结果 Linc00152在口腔鳞癌组织中的表达显著高于癌旁正常组织,在人口腔鳞状细胞癌细胞系TSCCA、SCCl5、CAL27和SCC9中的表达显著高于人正常黏膜HOK细胞,差异均有统计学意义(P<0.05);经相关性分析,Linc00152的高表达与淋巴结转移有关(P<0.05);CAL27和SCC9细胞系敲低Linc00152后,CAL27和SCC9细胞增殖、侵袭和转移明显受到抑制(P<0.05)。结论 linc00152在口腔鳞癌组织和口腔鳞癌细胞系中呈高表达,下调linc00152表达可抑制口腔鳞癌细胞增殖、侵袭和转移,linc00152可成为口腔鳞癌潜在治疗靶点。Objective To investigate the effect of long non-coding RNA Linc00152 on the proliferation and in-vasion of human oral squamous carcinoma (OSCC). Methods The expression of Linc00152 in OSCC tissues, adjacent tissues and OSCC cell lines was detected by real-time polymerase chain reaction (RT-qPCR). After CAL27 and SCC9 cells were transfected with Linc00152-siRNA, the proliferation, invasion and metastasis of CAL27 and SCC9 cells were detected by CCK8, scratch and Transwell test. Results The expression of Linc00152 in OSCC tissues was significantly higher than that in adjacent normal tissues (P〈0.05). The expression of Linc00152 in OSCC cell lines TSCCA, SCCl5, CAL27 and SCC9 was significantly higher than that in normal mucosal HOK cells (P〈0.05). The correlation analysis showed that the high expression of Linc00152 was related with lymph node metastasis (P〈0.05). After the knockdown of Linc00152 in CAL27 and SCC9 cell lines, the proliferation, invasion and metastasis of CAL27 and SCC9 cells were sig-nificantly inhibited (P〈0.05). Conclusion The expression of Linc00152 is high in OSCC tissues and OSCC cell lines, and down-regulation of the expression of Linc00152 can inhibit the proliferation, invasion and metastasis of OSCC cells. Linc00152 can be used as a potential therapeutic target for OSCC.
