摘 要:目的探讨茶多酚在脂多糖诱导的人支气管上皮细胞损伤中的保护作用及其机制。方法培养人支气管上皮细胞16HBE,给予不同浓度的脂多糖(1,25,50,100μg/ml)和茶多酚(100,200,400,800g/ml)作用。MTT检测细胞的生长,流式细胞术检测细胞凋亡,western blot检测细胞核内和总NFκB的蛋白表达,ELISA检测IL-6,IL-13和TNF-α的含量,试剂盒检测MDA,SOD和LDH的水平。结果与对照组相比,LPS组的细胞生长显著降低;细胞凋亡显著升高;IL-6,IL-13和TNF-α的含量升高;核内和NFκB总蛋白上升;MDA和LDH含量增加,SOD的活性降低,而茶多酚处理抑制以上LPS诱导的16HBE损伤。结论茶多酚可以通过抑制氧化应激缓解脂多糖诱导的人支气管上皮细胞损伤。Objective To investigate the effect of Tea Polyphenols(TP)on lipopolysaccharide(LPS)-induced human bronchial epithelial cell injury and underlying mechanism. Methods Cultured human bronchial epithelial cells(16HBE)were treated with LPS(1,25,50,100μg/ml)and TP(100,200,400,800g/ml),respectively.Cell proliferation was measured by MTT method.Cell apoptosis was detected by flow cytometry assay.The expression levels of nuclear NFκB and NFκB were measured by western blot.The content of IL-6,IL-13 and TNF-αwere detected by Elisa method.The MDA content,LDH content,SOD activity were measured by corresponding kit. Results Compared with the control group,the cell proliferation was significantly decreased;cell apoptosis was increased;the content of IL-6,IL-13,TNF-αwere increased;protein expression of nuclear NFκB,NFκB were increased;the content of MDA,LDH were increased,activity of SOD was decreased.LPS induced 16 HBE injury was improved by TP treatment. Conclusion tea polyphenols attenuates lipopolysaccharide-induced human bronchial epithelial cell injury through inhibiting oxidative stress.
