摘  要:目的探讨大鼠前额叶皮质miR-182表达与酒精依赖的关系,以及可能的作用机制。方法将60只大鼠随机分为持续饮酒组、戒断组和对照组,每组各20只。用灌胃法制备酒精依赖模型,用实时荧光定量PCR检测酒精依赖大鼠前额叶皮质miR-182与BDNFmRNA的表达,并对两者的表达量进行Pearson相关分析。结果三组间miR-182表达水平的差异有统计学意义(F=44.79,P〈0.01),两两比较差异均有统计学意义(P〈0.05)。持续饮酒组与对照组比较,BDNF的mRNA表达下调,差异有统计学意义(P〈0.05)。miR-182与BDNFmRNA表达量之间无相关性(P〉0.05)。结论酒精依赖大鼠前额叶皮质miR-182表达水平下调,miR-182可能参与酒精依赖的发病过程。Objective To study the relationship between expression of miR-182 in prefrontal cor- tex and alcohol dependence in rats and explore the possible mechanism. Methods Totals of 60 rats were randomly divided into 3 groups: continuous alcohol group (n=20), withdrawal group (n=20)and control group (n=20). Lavage was used to build alcohol dependence model. Real-time quantitative PCR was used to detect the expression of miR-182 and BDNF mRNA in prefrontal cortex of alcohol dependent rats. Pearson analysis was carried out to analyse the correlation between expression of miR-182 and BDNF mRNA. Results The expres- sion of miR-182 was significantly different among three groups(F=44.79, P 〈 0.01). Significant differences were found between each two groups (P 〈 0.05). Compared with control group, the expression of BDNF mRNA was significantly decreased in continuous alcohol group (P 〈 0.05). The expression of miR-182 and BDNF mRNA was not correlated (P 〈 0.05). Conclusions The expression of miR-182 in prefrontal cortex of alcohol depen- dent rats is decreased, miR-182 may be involoved in the pathogenesis of alcohol dependence.
