摘 要:为分析淀粉酶对金针菇(Flammulina velutipes)子实体发育的作用,针对已经鉴定的金针菇淀粉酶家族基因(6个α-淀粉酶和1个γ-淀粉酶),采用定量PCR方法对其在不同发育时期(从原基形成第1天到第12天成熟开伞期)、不同组织部位(菌盖和菌柄)的表达量进行测定,进而分析其表达模式。结果显示:7个淀粉酶基因在菌柄中表达量均高于菌盖;其中,α-Amy-5和γ-Amy-1在菌柄中的表达量变化为先下调再上调,两个基因预测编码蛋白均有信号肽,亚细胞定位在细胞外,推测其可能参与菌柄细胞壁的形态建成;α-Amy-3亚细胞定位在线粒体中,与其它各发育时期相比,其在菌柄快速伸长期表达量达到最高。α-Amy-1和α-Amy-6基因则在金针菇成熟期表达量最高,推测其可能与金针菇菌盖的发育有关。To analyze the role of amylases on the development of fruiting body in Flammulina velutipes,seven amylase family genes(including sixα-amylases and oneγ-amylase)identified in F.velutipes were analyzed in pileus and stipe at different developmental stages(D1 of primordial stage to D12 maturation stage)using quantitative PCR.The results showed that all seven amylase genes exhibited greater expression quantity in stipe than those in pileus.The relative expression level of amylaseα-Amy-5 andγ-Amy-1 in stipe appeared to be least at day 8 of the elongation period.Bothα-Amy-5 andγ-Amy-1 were predicted to contain extracellular targeting signal peptides,suggesting their involvement in stipe cell wall development.Amylaseα-AMY-3 was predicted to localize in mitochondria,and its expression level reached the highest in the rapid stipe elongation stage.Amylase α-Amy-1 and α-Amy-6 showed the greatest expression in pileus at maturation stage,suggesting their participation in the development of pielus in F.velutipes.
