生酮饮食联合抑制糖异生药物与肿瘤治疗

摘　　要：肿瘤是威胁人类健康和导致死亡的主要原因之一.目前常用于治疗肿瘤的方法包括手术、放疗、化疗以及生物靶向治疗等.虽然有较少部分的早期肿瘤已经可以治愈,但绝大部分肿瘤仍然缺乏有效的治疗方法.且上述治疗措施仅能延缓肿瘤患者的生存时间,达不到治愈肿瘤的效果.本研究致力于总结前人的研究成果,寻找治疗肿瘤的有效方法.Seyfried等提出"癌症是代谢性疾病"这一假说,即肿瘤的发生与线粒体功能受损有关,线粒体遭破坏,细胞能量缺乏,为了生存,细胞必须通过其它途径获得能量,肿瘤细胞转变成利用糖酵解进行供能.如果这一假说能够证实,肿瘤治疗就可以通过切断葡萄糖的供应,将饮食变为高质量脂肪、优质蛋白质、低碳水化合物的生酮饮食以及联合抑制糖异生的药物达到切断肿瘤细胞的能量供应进而"饿死"肿瘤细胞.该治疗方案能够起到"饿死"肿瘤细胞的作用,以前被认为是绝症的肿瘤将有希望得到有效地治疗,给肿瘤患者带来曙光.Tumor has become the first major factor threatening health in the world which can lead to death. At present,the methods include surgery,radiotherapy,chemotherapy and biological target therapy used to treat cancer. Less parts of early cancer can cure,but most of these tumors are lack of effective treatment methods.The above treatments can only delay the survival time of patients with tumor which fail to cure the tumor. The review summarizes the research achievements of predecessors and finds an effective method of tumor treatment. Seyfried, et al, put forward the hypothesis that cancer is a metabolic disease,which means the occurrence of tumor is associated with function impairment of mitochondria,when the mitochondria suffers damage,cells are lack of energy deficiency of cells,in order to survive,the cells must get energy through other way,then the tumor cells mainly use glycolysis to get energy. If this hypothesis is confirmed,we cut off the supply of glucose,eat the food into a high quality of fat,high protein and low carbohydrate and combine with inhibition of gluconeogenesis drugs at the same time,to cut off energy supply of the tumor cells,it can starve tumors cells. This treatment can play a role in starving tumor cells,and cancer previously considered as a terminal illness will hopefully get effective treatment,which will bring light to the tumor patients.
