摘 要:为了寻找与肝细胞癌转移相关的蛋白,本研究选择了具有相同遗传背景但不同转移能力的两个肝癌细胞系MHCC97H和HCCLM6,在采用超滤辅助样品制备(filter aided proteome preparation,FASP)方法和相对和绝对定量同位素标记(isobaric tags for relative and absolute quantitation,i TRAQ)技术对样品进行处理后,对所提取的蛋白质进行了质谱鉴定和定量分析,对差异蛋白进行了包括亚细胞定位,生物学过程和分子功能富集在内的生物信息学分析。我们共鉴定了4 987个蛋白,与MHCC97H细胞系相比HCCLM6中有93个差异表达蛋白(|ratio|≥1.5,一个样本t检验,p<0.05),其中40个蛋白上调,53个蛋白下调,且大多数差异表达的蛋白质位于细胞质和细胞膜。GO分析表明与细胞粘附和蛋白质结合相关的蛋白质在差异蛋白中显著富集。本研究为肝癌转移机制的进一步研究提供了一定的帮助。In this study,quantitative proteomics analysis was performed on hepatocellular carcinoma(HCC) cell lines with low and high metastatic potentials for discovery of candidate HCC metastasis-associated proteins.Two HCC cell lines(MHCC97H and HCCLM6),which have the same genetic background but different metastatic potentials,were chosen for analysis.Proteins extracted from cells were analyzed by MS identifi cation after FASP digestion and i TRAQ labeling.Bioinformatics an alysis of differentially expressed proteins,including subcellular localization,biology process and molecular function enrichment were performed.A total of 4 987 proteins were identified.There were 93 differentially expressed proteins(|ratio|≥1.5,one sample t test,p〈0.05),among which 40 proteins were upregulated and 53 proteins were down-regulated in HCCLM6.Most of the differentially expressed proteins were located in cytoplasm and cell membrane.GO analysis suggested that proteins related to cell adhesion and proteinbinding were significantly enriched.This study profiled the proteomes of HCC cell lines with different metastatic potentials,which provided valuable reference for future studies on HCC metastasis.
