摘 要:【目的】通过观察LE540对形觉剥夺性近视(form-deprivation myopia,FDM)豚鼠的屈光度、眼轴及巩膜细胞外基质(extracellular matrix,ECM)的影响,探讨LE540在FDM豚鼠巩膜中的作用。【方法】3周龄三色豚鼠24只随机分组,每组8只。Ⅰ组为形觉剥夺组,左眼用气球头套遮盖;Ⅱ组左眼球后注射0.4 g/L视黄酸(retinoic acid,RA),Ⅲ组行左眼遮盖,并注射0.1 g/L LE540,三组均饲养至第24天。右眼均为自身对照眼。实验前后均行视网膜检影测量屈光度,实验结束后取眼球测量眼轴长,取后极部巩膜组织,检测各组豚鼠后巩膜中基质金属蛋白酶-2(matrix metalloproteinase-2,MMP-2)和核心蛋白多糖(decorin,DCN)的蛋白表达。【结果】实验后Ⅰ组、Ⅱ组豚鼠左眼均形成近视;Ⅲ组左眼仍为远视,且三组间差异有统计学意义(H=10.38,P=0.006)。实验后Ⅰ组、Ⅱ组豚鼠左眼轴比右眼明显增长(P<0.05);Ⅲ组左右眼的眼轴无显著差异(P=0.146),双眼眼轴差值三组间比较有显著性差异(H=6.77,P=0.034)。实验后左眼的MMP-2/β-actin与DCN/β-actin三组比较差异有显著统计学意义(F=7.464,P=0.012;F=4.448,P=0.045),其中Ⅰ组和Ⅱ组无显著性差异,但与Ⅲ组比较均有显著性差异。【结论】形觉剥夺和外源注射视黄酸均可导致轴性近视,视黄酸诱导近视形成的作用可被LE540不完全抑制。【Objective】To observe the effect of LE540 on refractive state and extracellular matrix(ECM) of axial length and sclera in guinea pigs with form-deprivation myopia(FDM),and explore the role of LE540 in the sclera of FDM guinea pigs.【Methods】A total of24 pigmented guinea pigs aged 3 w were randomly divided into three groups.The left eyes of guinea pigs in the groupⅠ(n=8) were covered by facemasks for 24 d.The left eyes of guinea pigs were injected with 0.1 ml retinoic acid(RA)(0.4 g/L) every three days in the groupⅡ(n=8).The left eyes of guinea pigs in the group Ⅲ(n=8) were injected with 0.1 ml LE540(0.1 g/L) every three days and also covered with facemasks.All the guinea pigs were treated for 24 d.Right eyes were used as the self control eyes.Diopter was measured before and after the experiment with retinoscopy.Ocular axial length was measured with micrometer.The levels of matrix metalloproteinase-2(MMP-2) and decorin(DCN) were detected in all groups.【Results】Myopia was induced in the left eyes of the groups Ⅰand Ⅱ,but the left eyes were still hyperopia in the group Ⅲ,and there was statistically significant difference between the three groups(H=10.38,P=0.006).Axial length of left eyes were obviously longer than that of right eyes in the groups Ⅰand Ⅱ(P0.05).The difference of axial length between left and right eyes in the group Ⅲ was not significant(P=0.146).The binocular axial difference between the three groups was significantly different(H=6.77,P=0.034).The difference of MMP-2/β-actin and DCN/β-actin in left eyes between the three groups was statistically significant(F=7.464,P=0.012;F=4.448,P=0.045).There was no significant difference between group I and group II,but they were significantly different from group Ⅲ.【Conclusion】 Axial myopia can be induced by form deprivation and exogenous RA injection.The induction of FDM by RA may be partly inhibited by LE540.
