电刺激对脑缺血／再灌注大鼠脊髓背角细胞凋亡的影响

摘　　要：目的观察穴位电刺激对脑缺血，再灌注大鼠脊髓背角细胞凋亡的影响。方法采用Longa线栓法建立大鼠左侧大脑中动脉缺血，再灌注模型，将80只大鼠按随机数字表法分为对照组（C组）和电刺激组（E组）（每组40只）。E组予针刺患侧曲池、足三里穴位，C组不予干预。两组于造模后1d（T1）、3d（T2）、7d（T3）、14d（T4）4个时间点随机选取10只大鼠，应用TUNEL法检测脊髓背角细胞凋亡，免疫组织化学检测caspase-3蛋白表达。结果两组组内比较，脊髓背角细胞凋亡率及caspase-3蛋白表达阳性细胞率，T2、T3和T4时都比T1时高（P〈0．05或P〈0．01）；组间比较，E组L、T3、L时细胞凋亡率及caspase-3蛋白表达阳性细胞率均比C组降低，差异有统计学意义（P〈0．05），T1时两组细胞凋亡率及caspase-3蛋白表达阳性细胞率差异无统计学意义（尸〉0．05）。结论穴位电刺激可以降低脊髓背角细胞凋亡。Objective To observe the effects of electroacupuncture stimulation on the apoptosis of spinal dorsal horn cells in rats after cerebral ischemia/reperfusion. Methods A left cerebral artery occlusion/reperfusion model of rats was established by the Longa suture method. Then, a total of 80 rats were randomly divided into two groups （n =40）： a control group （group C） and an electroacupuncture group （group E）. Quchi and Zusanli acupoints on the affected side were acupunctured for group E, while no intervention was performed for group C. Then, 10 rats were selected randomly from each group 1, 3, 7, 14 d after modeling （T1, T2, T3 and T4）. Then, the apoptosis in spinal dorsal horn cells was detected by TUNEL method, and the level of caspase-3 was measrued by immunohistochemistry. Results Both groups E and C showed higher apoptotic rates in spinal dorsal horn ceils and positive rates of caspase-3 protein at T2, T3 and T4 than T1 （P〈0.05 or P〈0.01）. There was no difference in apoptotic rate at T1 between the two groups （P〉0.05）. Meanwhile, statistical difference was found as to apoptotic rate in spinal dorsal horn cells and positive rates of caspase-3 protein at T2, T3 and T4 between the two groups （P〈0.05）. Conclusions Electroacupuncture stimulation can reduce the apoptosis in spinal dorsal horn cells.
