摘 要:目的研究双氢青蒿素对中波紫外线照射后小鼠皮肤组织白介素(interleukin,IL)-1,IL-6及半胱氨酸天冬氨酸蛋白酶(cysteinylaspartic speahc proteinase)-3,Caspase-8表达的影响,初步探讨青蒿素光保护作用的可能机制。方法 40只BALB/C小鼠随机分为4组,即对照组(C),NB-UVB照射组(UVB),双氢青蒿素组(DHA),双氢青蒿素+NB-UVB组(DHA+UVB)。C组给予生理盐水灌胃,UVB组给予生理盐水灌胃后NB-UVB照射,DHA组给予双氢青蒿素灌胃,DHA+UVB组给予双氢青蒿素灌胃后进行NB-UVB照射。各组灌胃100mg/(kg·d),持续2周。UVB照射剂量为1 J/(cm^2·d),共2周,累积剂量14 J/cm^2,14天后取小鼠背部皮肤组织,采用酶联免疫吸附试验检测各组小鼠皮肤组织IL-1,IL-6水平,RT-PCR,WT-blot检测各组小鼠皮肤组织Caspase-3,Caspase-8变化情况。结果与C组相比,UVB组皮肤组织中IL-1,IL-6明显增加,Caspase-3,Caspase-8 mRNA表达及其蛋白水平均增加,差异均有统计学意义(均P<0.05),DHA组小鼠皮肤组织IL-1,IL-6水平没有明显变化;Caspase-3,Caspase-8表达降低,但差异均无统计学意义(均P>0.05)。DHA+UVB组与UVB组相比,IL-1,IL-6的浓度增加明显减少,Caspase-3,Caspase-8 mRNA表达及蛋白水平降低,差异均有统计学意义(均P<0.05)。结论双氢青蒿素对紫外线所致小鼠光损伤的炎症因子IL-1,IL-6及凋亡蛋白Caspase-3,Caspase-8的产生有一定的抑制作用。提示系统性应用双氢青蒿素对UVB致小鼠光损伤有保护作用。Objective To study the protective effect of dihydroartemisinin on chronic skin photodamage, and how it influcnces the expression of IL-1,IL-6,Caspase-3,Caspase-8 on mouse skin induced by UVB.Methods Forty BALB/C mouse were randomized into four groups, namely,comprising control group(C),ultraviolet exposed group(UVB),dihydroartemisinin injection-treated group(DHA)and dihydroartemisinin injection-treated and ultraviolet exposed group(DHA+UVB).Mouse of the normal control group were injected with normal sodium; Mouse of UVB group were irradiated with NB-UVB radiation(after the injection of normal sodium); DHA group were injected with dihydroartemisinin and DHA+UVB group were treated with dihydroartemisinin injection and ultraviolet exposure.A dose of 100 mg/kg and a dose of 1 J/cm2 was given every day for 2 weeks.After NB-UVB radiation,the skin specimens were obtained.ELISA method was used to detect the Caspase-3 and Caspase-8 protein expression in these specimens.Results Compared with the c group,there was a significant increase in the expression of IL-1,IL-6,Caspease-3,Caspease-8 in UVB and DHA+UVB group(all P〈0.05),but compared with the UVB group,IL-1,IL-6,Caspease-3,Caspase-8 expression levels in the DHA+UVB group decline( all P〈0.05).Conclusion Our results suggest that dihydroartemisinin can protect against the damage induced by UVB in mouse skin,and IL-1,IL-6,Caspease-3,Caspase-8 maybe play a role.
