摘 要:目的:探讨Lgr5基因在新疆维、汉族直肠癌组织和癌旁组织中的表达及其与临床病理因素的关系。方法:收集我院2014年1月至2016年3月期间住院的Ⅱ、Ⅲ期准备行术前放化疗的直肠癌患者肠镜活检组织,癌组织及对应癌旁组织各94例,其中汉族50例,维吾尔族44例,应用免疫组织化学(SP)法和实时荧光(qRT-PCR)法分别检测直肠癌组织和癌旁组织中Lgr5蛋白和mRNA的表达。结果:直肠癌组织中Lgr5蛋白的阳性表达率为85.1%,高于癌旁组织的24.5%,差异有统计学意义(P<0.05);Lgr5 mRNA表达水平在癌组织中(9.77±7.04)高于癌旁组织(3.25±2.89),差异有统计学意义(P<0.05);维吾尔族、汉族Lgr5蛋白和mRNA表达在癌组织和癌旁组织中均无统计学差异(P>0.05)。Lgr5蛋白表达与TNM分期和浸润深度有关,Lgr5 mRNA表达与组织分化程度和淋巴结转移有关。结论:Lgr5基因表达与直肠癌的发生、发展有相关性;Lgr5基因表达在维、汉不同民族之间无显著性差异。Objective:To detect the expression of Lgr5 in rectal cancer tissues and adjacent tissues and its correla- tion with clinical factors in Xinjiang Uygur and Han ethnic groups. Methods:We collected clinical stage II -III rectal cancer pathological tissue by colonoscopy between January 2014 and March 2016 in our hospital. Expression of Lgr5 ( leucine - rich repeat - containing G - protein - coupled receptor5 ) protein and mRNA were respectively assessed in 94 cases of cancer tissue and 94 cases of adjacent tissue by immunohistochemistry and real time fluorescence qRT - PCR method. Results :The positive expression rate of Lgr5 protein in rectal cancer was significantly higher than that in the normal tissues (85.1% vs 24.5 % ,P 〈 0.05). The expression of Lgr5 mRNA in cancer tissues (9.77 ± 7.04) was higher than that in normal tissues (3.25 ± 2.89), which was statistically significant (P 〈 0.05 ). There were no differences in expression of Lgr5 protein and mRNA in cancer tissues and adjacent tissues in the Uygur and Han na- tionalities( P 〉 0.05 ). The Lgr5 protein expression was related to the TNM staging and the depth of invasion, Lgr5 mRNA expression was related to the degree of tissue differentiation and lymph node metastasis. Conclusion: The Lgr5 gene expression were related to the rectal cancer occurrence, development. The expression of Lgr5 gene in the Xinjiang Uygnr and Han rectal cancer patients was not different.
