摘 要:目的评估比较双重固定法与传统甲醛固定法对骨髓活检标本固定效果的差别。方法采用传统甲醛固定法与双重固定法分别对70例标本进行固定,再经常规的脱钙、脱水、浸透、石蜡包埋和切片后进行HE染色,并对两种固定方法的最终结果进行观察。结果经中性甲醛固定的组织细胞收缩明显,结构模糊,颜色暗淡,而双重固定法固定的细胞形态舒展,色泽艳丽,胞核及胞浆清晰,组织结构保存完整。结论双重固定法较传统甲醛固定法,标本固定效果显著提升,为临床疾病的诊断提供了更好的直观依据。Objective To evaluate the difference of the fixation effect between the two-step fix- ation method and the traditional formaldehyde fixation method in bone marrow biopsy. Methods A total of 70 specimens were fixed by the traditional neutral formaldehyde fixation and double fixation method, and then by normal decalcification, dehydration, soaking, paraffi and HE staining, at last the cell morphological results were observed. Results n embedding The tissues fixed by neutral formaldehyde were shrinkage, blurred and dim, while fixed by two-step fixation were stretch, colorful, clearly and completely. Conclusion The effect of two-step fixation method is better than the traditional neutral formaldehyde.
