摘 要:目的:基于Wnt信号通路观察补肾益脑灸法对围绝经期抑郁症大鼠海马神经干细胞分化标记物表达的影响并探讨其治疗机制。方法:60只大鼠按随机数字表法分为空白组、模型组、假手术组、西药组和艾灸组5组,采用去势和慢性不可预见刺激结合制备围绝经期抑郁症大鼠模型。连续治疗28 d,观察大鼠一般情况变化、动情周期、体质量及糖水消耗实验结果,运用ELISA法检测海马组织中的GFAP、Tuj-1、GSK-3β、β-catenin,RT-PCR法检测海马组织中的GSK-3βmRNA、β-catenin mRNA表达。结果:造模后其他组较空白组表现出抑郁行为,而治疗后抑郁行为好转,西药组、艾灸组及假手术组较模型组海马组织中GFAP、GSK-3β及GSK-3βmRNA的表达量增加,Tuj-1、β-catenin及β-catenin mRNA表达量有所减少。结论:补肾益脑灸法可以通过调节海马Wnt通路GSK-3β、β-catenin的表达调控海马神经干细胞的定向分化,对围绝经期抑郁症起到治疗作用。Objective: To observe the expression of neural stem cell differentiation markers on Wnt signaling pathway in perimenopausal depressive rat 's hippocampus; to explore the mechanism of moxibustion treatment for nourishing"Kidney-Brain System". Methods: 60 rats were randomly divided into five groups: a normal group,a model group,a sham operation group,a western medicine group,and a suspended moxibustion( SM) group. A perimenopausal depressive rat's model was made by castration and chronic unpredictable stress combination. The treatments were for 28 days. The general condition of rats,the rat 's estrous cycle,the weight of rats,and sugar consumption experiment of rats were observed. GFAP,Tuj-1,GSK-3 beta and beta-catenin in rat 's hippocampus tissue were detected by Elisa. RT-PCR method was used to detect the expression of GSK-3βmRNA and β-catenin mRNA in the rat's hippocampus. Results: After modeling,except the normal group,the depressive behavior of all other groups was showed,however,after treatment,the depressive behavior of rats was dramatically reduced. Compared with the model group,the expression of GSK-3,GSK-3,mRNA and GFAP in the rat's hippocampus of western medicine group,moxibustion group and sham operation group was increased,Tuj-1,beta-catenin and beta mRNA-catenin expression was reduced. Conclusion: Moxibustion treatment could nourish "Kidney-Brain System"through changing the expression of GSK-3β and β-catenin on Wnt signaling pathway in perimenopausal depressive rat's hippocampal and through regulating the directional differentiation of neural stem cells in the hippocampus. Moxibustion has effect on the treatment of perimenopausal depression.
