摘 要:目的:检测miRNA-873在肝细胞肝癌组织以及细胞系中的表达情况,并进一步探讨miRNA-873对肝癌细胞迁移和侵袭的影响。方法:实时定量逆转录聚合酶链反应(RT-PCR)检测肝细胞肝癌组织、癌旁组织以及细胞系中miRNA-873的表达水平。分别用划痕实验和Transwell细胞侵袭实验检测细胞迁移和侵袭能力。蛋白质印迹(Western blot)检测Janus激酶2(Janus kinase 2,JAK2)和信号转导子与转录活化子3(signal transduction and transcriptional activation 3,STAT3)活性的变化。结果:肝癌组织和肝癌细胞系中miRNA-873表达水平明显高于对应的癌旁组织和正常肝脏细胞。RT-PCR证实转染miRNA-873 inhibitor或mimics后miRNA-873的表达量明显降低或升高(P<0.01)。与对照组(NC)相比,敲低miRNA-873明显抑制肝癌细胞HepG2迁移和侵袭,差异有统计学意义(P<0.05)。过表达miRNA-873则明显促进肝癌细胞Huh7迁移和侵袭,差异有统计学意义(P<0.05)。miRNA-873可明显活化JAK2/STAT3信号通路。结论:miRNA-873在肝癌中高表达,且可能通过活化JAK2/STAT3信号通路促进肝癌的迁移与侵袭。Objective: To investigate the expression of miRNA-873 in hepatocellular carcinoma (HCC) tissues and cell lines,further study the effect of miRNA-873 on migration and invasion in hepatocellular carcinoma cell lines.Methods: The expression of miRNA-873 in HCC tissues and cell lines was detected by RT-PCR.Wound-healing experiment and Transwell invasion were used to examine migration and invasion of cells,respectively.Western blot was used to detect the activity of Janus kinase 2 (JAK2) and signal transduction and transcriptional activation 3 (STAT3).Results: The results showed that miRNA-873 was upregulated in HCC tissues and HCC cell lines,compared with that in the normal control.RT-PCR showed that the expression of miRNA-873 decreased or increased after miRNA-873 inhibitor or mimic transfection,respectively (P〈0.01).Compared with the normal control,knockdown of mi R-873 significantly inhibited the migration and invasion of HepG2 (P〈0.05).However,overexpression of miRNA-873 had the opposite effect (P〈0.05).We also found that miRNA-873 significantly activated JAK2/STAT3 signal pathway.Conclusion: Mi RNA-873 was overexpressed in HCC and might promote HCC migration and invasion through JAK2/STAT3 signal pathway.
