摘  要:目的观察运动训练对缺血性卒中后抑郁(PSD)大鼠的干预作用。方法将50只雄性SD大鼠随机均分为假手术组、卒中组、抑郁组、PSD组和运动组。卒中组采用大脑中动脉栓塞法制备局灶性脑缺血大鼠模型,抑郁组采用孤养结合慢性不可预见性温和刺激制备抑郁大鼠模型,PSD组将以上方法相结合制备PSD大鼠模型,运动组制备PSD大鼠并同时进行4周跑台训练。28d后测定五组大鼠体重、糖水消耗量、旷场实验(OFT)行为学评分,采用高效液相色谱法测定五组大鼠脑内5-羟色胺(5-HT)含量。结果 28d后,PSD组和抑郁组体重、糖水消耗量、OFT评分均低于假手术组(P<0.01或P<0.05);卒中组、抑郁组和运动组大鼠体重、糖水消耗量和OFT评分均较PSD组增加(P<0.01或P<0.05)。与假手术组比较,抑郁组、PSD组大鼠海马和大脑皮质5-HT含量减少(P<0.01);卒中组、抑郁组、运动组大鼠海马和大脑皮质5-HT含量较PSD组增加(P<0.01)。结论运动训练可以改善PSD大鼠的行为,提高大鼠脑内5-HT含量。Objective To observe the intervention effect of exercise training on the ischemic post-stroke depression(PSD)rats.Methods Fifty male SD rats were randomly divided into five groups with 10 rats each.Group A was taken as sham operation.The focal cerebral ischemia rat model was established by middle cerebral artery occlusion in group B.The depression rat model was established by the solitary combined with chronic unpredictable mild stimulation in group C.The PSD rat model was established by combining the above methods in group D.The PSD rat model was established and received treadmill training for 4 weeks in group E.After 28 days,the weight,sugar water consumption and open field test(OFT)score were detected.The level of 5-hydroxytryptamine(5-HT)was assayed by high performance liquid chromatography.Results Compared with group A,the weight,sugar water consumption and OFT score were decreased in groups of C and D,which were higher in groups of B,C and E than those in group D(P〈0.05 or P〈0.01).Compared with group A,the level of 5-HT in the hippocampus and cerebral cortex was lower in groups of C and D,which was decreased in group D than that in groups of B,C and E(P〈0.01).Conclusion Exercise training can improve the behavior of PSD rats and increase the level of 5-HT in brain.
